摘    要:目的 基于网络药理学探讨当归四逆汤治疗子宫内膜异位症(EMS)的作用机制。方法 利用中药系统药理学数据库与分析平台(TCMSP)筛选当归四逆汤有效活性成分及潜在靶点,在Genecard、OMIM和TTD数据库中寻找EMS相关靶点基因取当归四逆汤靶点基因与EMS相关靶点基因共同靶点。通过Cytoscape 3.5.0软件构建“当归四逆汤活性成分-潜在靶点-EMS”网络图。运用String数据库构建构建蛋白质互作(PPI)网络图。对潜在靶点基因进行基因本体(GO)富集分析和京都基因和基因组百科全书(KEGG)通路分析。结果 数据库筛选的得到当归四逆汤有效活性成分151个,与EMS相关潜在作用靶基因71个,KEGG富集通路分析主要包括化学致癌受体激活,脂质和动脉粥样硬化通路等。结论 当归四逆汤具有多活性成分,能通过多靶点,多通路发挥EMS保护作用。

Abstract:Objective To investigate the mechanism of Danggui Sini Decoction in the treatment of endometriosis (EMS) based on network pharmacology. Methods The traditional Chinese medicine system pharmacology database and analysis platform (TCMSP) was used to screen the active ingredients and potential targets of Danggui Sini Decoction, and EMS-related target genes were searched in Genecard, OMIM and TTD databases, and the target genes of Danggui Sini Decoction and EMS were selected. Common target of related target genes. The network diagram of "active components of Danggui Sini Decoction-potential targets-EMS" was constructed by Cytoscape 3.5.0 software. The protein interaction (PPI) network map was constructed using the String database. Gene ontology (GO) enrichment analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis were performed on potential target genes. Results The database screened 151 active ingredients of Danggui Sini Decoction and 71 potential target genes related to EMS. The KEGG enrichment pathway analysis mainly included chemical carcinogenesis-receptor activation, lipid and atherosclerosis pathways. Conclusion Danggui Sini Decoction has multiple active components, which can exert EMS protection through multiple targets and multiple pathways. Animal experiments confirmed that Danggui Sini Decoction can improve EMS.
