摘    要:目的比较两种SD大鼠多囊卵巢综合征动物模型的建模效果。方法建立两种PCOS动物模型。观察各组大鼠卵巢形态学、性激素及空腹血糖和胰岛素的变化。结果DHEA建模组大鼠的体重显著低于对照组(P〈0.05);hCG建模组大鼠卵巢重量和体积均显著高于对照组(P〈0.05)。两建模组大鼠卵巢卵泡多呈囊性扩张,颗粒细胞层数减少,卵泡膜细胞、间质细胞增生。两建模组大鼠血清T、FINS、FBG和HOMA指数均显著高于对照组(P〈0.05);DHEA建模组LH水平较对照组无显著性差异;hCG建模组大鼠血清LH、LH/FSH比值均显著高于对照组(P〈0.05)。DHEA建模组FSH水平显著高于对照组(P〈0.05)。结论利用hCG诱导SD幼年雌性大鼠出现了与PCOS患者极为相似的卵巢病理改变及LH、雄激素水平升高和胰岛素抵抗等典型内分泌变化,其建模效果优于DHEA。

Abstract:Objective: To compare the outcomes in two polycystic ovarian syndrome (PCOS) rat models. Methods: Two different Sprague -Dawley (SD) rat PCOS models were established. Ovarian morphologic changes, sex hormone levels, fasting blood insulin and glucose levels were detected. Results: The ovarian weight in DHEA induced PCOS group were lower than that in control group ( P 〈 0. 05 ), but the ovarian weight and volume in hCG induced PCOS group were both higher than those in control group ( P 〈 0. 05 ). The ovaries in both experimental groups showed multiple follicular cysts, and the number of theca cells and interstitial cells increased while the number of granulose cells decreased. The serum testosterone, fasting insulin, fasting blood glucose levels and the HOMA index were significantly higher in experimental groups than those in control groups ( P 〈 0. 05 ). There were no obvious differences in the serum LH level between DHEA induced PCOS group and the control group ( P 〉 0. 05 ). However, the serum LH level and the LH/FSH ratio were significantly higher in hCG induced PCOS group than those in control group ( P 〈 0. 05). The serum FSH level was significantly higher in DHEA induced PCOS group than that in control group ( P 〈0. 05) Conclusion: The rat model induced by hCG was proved to be an ideal PCOS animal model with more similar morphological and hormonal changes to those of PCOS patients than those induced by DHEA.
