胰岛素抵抗大鼠葡萄糖-6-磷酸酶的活性及基因表达变化

摘    要:目的:探讨高脂饮食致胰岛素抵抗动物模型中,葡萄糖-6-磷酸酶(G-6-Pase)的活性和基因表达改变与胰岛素抵抗的关系。方法:Wistar大鼠16只,随机分为2组,分别给予普通饲料和高脂饲料喂养,制作胰岛素抵抗模型。6周后测定各组体重、血糖、血脂、胰岛素变化,并利用高胰岛素-正常葡萄糖钳夹术评价胰岛素敏感性。测定肝脏微粒体G-6-Pase的活性和基因表达。结果:高脂饲料组大鼠体重、甘油三酯、游离脂肪酸和胰岛素水平显著增高,G-6-Pase活性增高[(0.51±0.11)vs(0.32±0.11)μmol/(min·mgprot),P<0.05],两组间的G-6-Pase基因表达无差异。结论:G-6-Pase参与了肝胰岛素抵抗的发病机制。

Abstract:Objective To explore the activities and gene expressions of glucose-6-phosphatase in fat-fed rats of insulin resistance. Methods sixteen rats were randomly divided into two groups(each group 8 rats). One group was given standard food and the other group was given high-fat diet to make insulin resistance model for 6 weeks. Insulin sensitivity was valued by hyperinsulinemic euglycemic clamp.
