弓形虫膜抗原ELISA检测抗弓形虫IgA抗体的方法及其临床应用的研究

摘    要:目的建立弓形虫膜抗原ELISA检测抗弓形虫IgA抗体的方法,并评价其临床应用价值。方法确定ELISA的最适反应条件。应用该法检测36例先天性、32例近期和30例慢性弓形虫感染者及15例连续观察的弓形虫病人治疗前后的血清IgA抗体。结果36例先天性、32例近期和30例慢性感染血清的IgA抗体检出数分别为26例、21例和0例。15例连续观察对象有8例在治后2月,6例在治后4月、1例在治后6月,其IgA抗体全部阴转。结论弓形虫膜抗原ELISA检测IgA抗体方法具有较高的敏感性、特异性和可重复性。IgA抗体的检测可应用于先天性和近期获得性弓形虫感染的早期诊断,且尚具有良好的疗效考核价值。

Abstract:Objective To establish methods of detection of Toxoplasma IgA antibodies a-gainst specific surface membrane antigen,and evaluate its clinical application. Methods To define the most suitable reaction condition for detection of specific IgA antibodies using surface membrane antigen of Toxoplasma by ELISA. The IgA antibodies were detected in 36 cases of congenital infect-ed melformation newborns, 32 recently and 30 chronically acquired infection cases, and 15 patients of toxoplasmosis who were continually observed before and after treatment. Results Among 36congenital infected malformation newborns, 32 recently acquired toxoplasmosis,the positive of IgA antibodies were 26 and 21 respectively. None of 30 individuals with chronic Toxoplasma infection had IgA antibodies. In 2, 4, 6 months after treatment, among 15 cases with toxoplasmosis, 8, 6, 1patients' IgA antibodies were negative respctively. Conclusion The method of detection of Toxo-plasma IgA antibodies against surface membrane antigen by ELISA is relatively high sensitive,spe-cific and it can be repeated. The method can be applied for early diagnosis of the congenital infection and recently acquired infection, and also can be used for evaluation of treatment.
