摘 要: | 目的探讨金顶侧耳提取物拮抗环磷酰胺致小鼠肝损伤的作用。方法小鼠以环磷酰胺(85 mg/kg×2)腹腔注射制备肝损伤模型,金顶侧耳提取醇沉上清部分按1.6,0.8,0.4 g/(kg.d)分为3个剂量组,沉淀部分按0.8,0.4,0.2 g/(kg.d)分为3个剂量组,腹腔注射给药(正常对照组注射生理盐水)13 d后测定各组小鼠血清中丙氨酸氨基转移酶(ALT)、门冬氨酸氨基转移酶(AST)的活性和丙二醛(MDA)的含量及肝组织MDA含量。结果与正常对照组比较,模型组小鼠ALT、AST和MDA含量明显提高(P均<0.05);与模型组比较,金顶侧耳提取物沉淀部分0.8 g/(kg.d)组和0.4 g/(kg.d)组小鼠ALT和AST的活性明显降低(P<0.05或0.01);上清部分0.4 g/(kg.d)组小鼠血清AST的活性明显降低(P<0.05);金顶侧耳提取物沉淀部分0.4 g/(kg.d)组和上清部分0.4 g/(kg.d)组小鼠血清MDA含量明显降低(P均<0.05)。结论金顶侧耳提取物具有一定的拮抗环磷酰胺致小鼠肝损伤的作用,其中沉淀部分的生物活性较强。

Study of protection of extracts from Pleurotus citrinipileatus Sing on liver injury induced by Cyclophosphamide in mice |

Abstract: | Objective It is to explore the protection of extracts from Pleurotus citrinopileatus on liver injury induced by Cyclophosphamide in mice.Methods The mice were given Cyclophosphamide by intraperitoneal injection to establish liver injury model.The supernatant in part of Pleurotus citrinopileatus alcohol extract was divided into three dose groups of 1.6,0.8,0.4 g/(kg·d),the precipitated in the part was divided into three dose groups of 0.8,0.4,0.2 g/(kg·d). 13 days after intraperitoneal injection(the control group was given normal saline),activities of serum ALT and AST and the content of MDA in serum and liver tissue were determined.Results Compared with the control group,the contents of ALT,AST and MDA in model group were significantly higher(P all<0.05).Compared with model group,the activity of AST was obviously decreased in the group which was given supernatant in part of Pleurotus citrinopileatus alcohol extract at the dosage of 0.4 g/(kg·d)(P <0.05),the content of serum MDA in the groups which were given supernatant or precipitation in part of the extract at the dosage of 0.4 g/(kg·d)(P all<0.05).Conclusion Pleurotus citrinopileatus extract has a certain antagonism on Cyclophosphamide-induced liver injury in mice,and the precipitation part has a stronger biological activity. |
