| 摘 要： | 目的通过构建RNAi表达载体，转染喉癌细胞株Hep 2，观察其对survivin表达的抑制作用。方法构建针对survivin基因的干扰载体，用脂质体转染喉癌细胞Hep 2，分析干扰载体对survivin表达的抑制情况，检测细胞凋亡。结果survivin在喉癌细胞Hep 2中呈高表达，其基因序列与GenBank序列对比一致，干扰载体pGensil/survivin能抑制survivin的表达，抑制率为68%，并诱导了喉癌细胞的凋亡。结论针对survivin的小分子RNA可以有效抑制喉癌细胞Hep 2中survivin蛋白的表达，并促进喉癌细胞凋亡。
