维生素E及其联合维生素C对老龄小鼠肝细胞凋亡的影响

摘    要:目的:观察维生素E(VE)及其联合维生素C(VE+ VC)的抗氧化作用、对肝细胞凋亡与增殖的调节作用以及抗氧化过程与细胞凋亡和增殖的关系。方法:昆明鼠随机分为幼龄组(4 月龄)、老年组(17 月龄)包括老年对照组、VE组、VE+ VC组;以硫代巴比妥酸法和磷钨酸法分别检测肝及血清过氧化脂质(LPO),流式细胞术测定肝细胞凋亡率并分析肝细胞周期。结果:老龄组肝及血清LPO含量均明显高于幼龄组,补以VE、VE+ VC后LPO含量明显降低,尤以VE+ VC效果明显;小鼠肝细胞凋亡率随月龄增加而明显升高,老龄组补以VE、VE+ VC后肝细胞凋亡率明显降低;与幼龄组相比,老龄组肝细胞发生G1 期阻滞,施以抗氧化处理后,G0/G1、S、G2+ M 各期细胞数均发生逆转性变化。结论:VE具有明显的抗氧化作用,与VC联用效果更佳,并可显著降低肝细胞凋亡的发生率,提高肝细胞增殖活力。

Abstract:In this study we detected the anti oxidative status, the changes in hepatocyte cycle and apoptosis and the relationship between them after treatment of VE,VE VC. Methods:Kunming mice were randomly grouped (12 mice in each group). VE(50mg/kg),VE VC(50mg/kg 100mg/kg) were given to each experimental group respectively. LPO was measured and apoptosis, cell cycle were analyzed by flow cytometry (FCM). Results:Compared with those in young gruop, the LPO of liver and serum increased significantly in elderly group, which could be decreased by VE, VE VC(P<0.05); with the development of senescence, apoptotic death increased in hepatocytes while after treatment of VE,VE VC the higher apoptotic rate decreased; FCM showed that the number of hepatocytes in the G 0/G 1 phase were increased, while those in S, G 2 M phases decreased in the elderly group as compared with young group. All the changes in proportion of hepatocytes in different phases could be reversed by VE,VE VC.Conclusion:VE posseses clear antioxidation. Its effect is better when combined with VC. They reduce the apoptotic rate in liver and enhance the proliferation of hepatocyte significantly.
