炎性因子对C2C12细胞、MC-3T3-E1细胞成骨、成肌相关基因表达的影响

摘    要:目的:探讨炎性因子对成肌和成骨细胞成肌、成骨相关基因表达的影响。方法:小鼠腹腔巨噬细胞上清液及白介素(IL)1β处理C2C12细胞及MC-3T3-E1细胞,观察炎性因子对两种细胞成骨、成肌相关基因的表达的影响。结果:小鼠腹腔巨噬细胞上清液促进C2C12细胞MyoD、Myogenin的表达,差别均有统计学意义(P〈0.001);促进MC-3T3-E1细胞碱性磷酸酶(ALP)的表达,吸光度比值上升为原来的3倍多,差别有统计学意义(P〈0.001)。IL-1β单独对C2C12细胞和MC-3T3-E1细胞的成肌、成骨因子的表达影响无统计学意义(P〉0.05)。结论:巨噬细胞上清液可上调MC-3T3-E1细胞的成骨相关基因表达;上调C2C12细胞的成肌相关基因表达。IL-1β单独不能对C2C12细胞和MC-3T3-E1细胞的成肌、成骨基因的表达产生影响。

Abstract:Objective: To explore the effect of inflammatory factors on the ossification and myogenesis related gene expressions in myoblasts and osteoblasts. Methods: C2C12 and MC-3T3-E1 cells were treated with supernatant of mice peritoneal maerophage and IL-1β. The ossification and myogenesis related gene expression in C2C12 and MC-3T3-E1 cells were examined. Results: Supernatant of mice peritoneal maerophage significantly up-regulated the MyoD and Myogenin expressions in C2C12 cells(P〈0.001) and ALP in MC-3T3-E1 cells (P〈0.001). IL-1β alone had no effect on the gene expression of ossification and myogenesis in C2C12 and MC-3T3-E1 ceils (P〉0.05). Conclusion: Supernatant of mice peritoneal macrophage could up-regulate myogenesis expression in C2C12 cells and ossification expression in MC-3T3-E 1 cells;IL-1 alone had no effect on the gene expression of ossification and myogenesis in C2C12 and MC-3T3-E1 ceils.
