参附注射液对小鼠移植心脏细胞粘附分子表达强度的影响

摘    要:目的通过检测小鼠移植心脏的细胞粘附分子,探讨参附注射液对移植心脏缺血再灌注损伤的保护作用。方法40只同基因近交系Balb/c小鼠,随机分为参附组与对照组,每组雌雄各10只,进行相同性别间配对移植,参附组于移植前连续3d每24h经腹腔给予参附注射液,最后1次给药为移植前30min;而对照组同法给予等量0,9%氯化钠注射液。分别于移植后4、24、72h及1周时对受体及移植心脏的成活率及移植物的形态进行观察,并通过免疫组织化学方法进行细胞内粘附分子-1(ICAM—1)、血管细胞粘附分子-1(VCAM—1)、血小板内皮细胞粘附分子(PECAM)、淋巴细胞功能相关抗原-1(LFA-1)染色,比较两组上述粘附分子的表达强度及LFA-1阳性细胞率的差别。结果参附组ICAM-1、VCAM-1的表达强度在移植后各时间点明显低于对照组(均P〈0.05);PECAM的表达强度未见明显差别;LFA-1阳性细胞率均低于对照组(P〈0.05)。参附组内各时间点比较,ICAM-1、VCAM-1、PECAM的表达强度和LFA-1阳性细胞率均在24h最高。结论参附注射液可有效抑制小鼠心脏移植后移植物内ICAM-1、VCAM-1的表达,减少LFA-1阳性细胞的浸润。

Abstract:Objective To investigate the protective effects of Shenfu injection against ischemic-reperfusion (IR) injury in murine cardiac transplantation. Methods Cardiac transplantations were performed between MHC fully-matched inbred mice. The animals were given Shenfu infection (Shenfu group) or normal saline (control group). Heart grafts were harvested at 4hr, 24hr, 72hr and 1 week after transplantation. H.E. staining and immunohistochemistry were performed to evaluate the morphology of the grafts and the expression of ICAM-1, PECAM, VCAM-1, LFA-1 molecules. Results In Shenfu group the expression of ICAM-1 and VCAM-1 was weaker than that of control group, particularly in the early stage after IR. No difference was found in PECAM expression at any time point between 2 groups. In Shenfu group, the rate of LFA-1 positive cells was less than that of control group and most significantly at 24 hrs. Conclusion Shenfu Injection can inhibit the expression of ICAM-1 and VCAM-1 and reduce the infiltration of LFA-1 positive cells in murine cardiac grafts
