基质金属蛋白酶-1和基质金属蛋白酶-2在非小细胞肺癌组织中的表达及临床意义

摘    要:目的:研究基质金属蛋白酶-1(MMP-1)和基质金属蛋白酶-2(MMP-2)在非小细胞肺癌(NSCLC)组织中的表达及临床意义。方法:应用免疫组织化学方法和Westernblotting检测70例非小细胞肺癌组织中MMP-1和MMP-2蛋白的表达,并进行图像分析测定电泳条带的吸光度A值。结果:70例非小细胞肺癌和30例正常肺组织中MMP-1、MMP-2表达阳性的例数分别为47.1%(33/70)、65.7%(46/70)和23.3%(7/30)、16.7%(5/30),差别具有显著意义。期肺癌MMP-1、MMP-2的平均吸光度A值明显高于、期。有淋巴结转移癌的MMP-1、MMP-2平均吸光度A值明显高于无淋巴结转移的病例,差异具有显著性意义。MMP-1在肺腺癌和鳞癌中的表达无显著性差异(P>0.05)。肺腺癌中MMP-2平均吸光度A值高于肺鳞癌,差异具有显著性意义(P<0.05)。结论:MMP-1、MMP-2在非小细胞肺癌组织中的表达增多,可能与非小细胞肺癌分期、淋巴结转移有关。

Abstract:Objective:To explore the expression level of MMP-1, MMP-2 in patients with primary non-small cell lung cancer (NSCLC). Methods:The expression of MMP-1, MMP-2 protein was detected in 70 NSCLC tissues by immunochemistry and Western blotting followed by image analysis in order to detect the A value of expression. Results:The positive rate of MMP-1 , MMP-2 protein expression in NSCLC tissues was 47.1% (33/70), 65.7%(46/70), higher than those in normal lung tissue.. The A value of MMP-1 , MMP-2 expression in NSCLC tissues of Ⅲ stage was significantly higher than that in NSCLC tissues ofⅠ,Ⅱstages (P<0.05). The A value of MMP-1, MMP-2 expression in NSCLC tissues of N1,2,3 groups was significantly higher than those in NSCLC tissues of N0 group (P<0.05).There was no significant difference in MMP-1 expression between NSCLC tissues of squamous cell carcinoma and adenocarcinoma(P>0.05). MMP-2 expression in NSCLC tissues of adenocarcinoma was significantly higher than that in NSCLC tissues of squamous cell carcinoma. Conclusion:MMP-1, MMP-2 expression was related to lymph node metastasis and TNM stages of lung cancer. MMP-1, MMP-2 protein expression may be associated with the invasion and progression of NSCLC
