毒素清颗粒对支原体肺炎老龄大鼠免疫功能和细胞因子的影响

摘    要:目的:探讨毒素清颗粒对支原体肺炎老龄大鼠免疫功能和细胞因子的影响。方法:SD大鼠随机分为青年对照组、青年模型组,老龄对照组、老龄模型组、毒素清组和阿奇霉素组,采用气管插管法复制支原体肺炎模型。分别以毒素清(3.995 g.kg-1.d-1)、阿奇霉素(23 mg.kg-1.d-1)灌胃7 d。第8天取材,检测外周血白细胞(WBC)和中性粒细胞(NEU)计数,支气管肺泡灌洗液(BALF)支原体PCR和分泌性免疫球蛋白(sIg)A、纤维连接蛋白(FN)表达,肺脏含水量和病理,血清肿瘤坏死因子(TNF)-α、白介素(IL)-6、IL-8表达。结果:阿奇霉素组BALF肺炎支原体阳性为0例(0%),毒素清组阳性3例(30%)。青年和老龄模型组BALF中sIgA和FN分别较青年、老龄对照组显著降低(P<0.01),肺组织含水量、外周血TNF-α,IL-6,IL-8显著升高(P<0.01),且较青年模型组变化显著(P<0.01)。与老龄模型组比较,毒素清组和阿奇霉素组sIgA,FN显著升高(P<0.01),肺组织含水量、TNF-α,IL-6,IL-8显著降低(P<0.01);毒素清组BALF中sIgA明显高于阿奇霉素组(P<0.01),IL-8低于阿奇霉素组(P<0.05)。结论:毒素清对老龄大鼠支原体肺炎作用明显,其机制可能与增强免疫功能、降低炎症因子表达,减轻炎症反应有关。
