藿香正气液对湿阻证大鼠抗氧化作用及对胃黏膜EGFR表达的影响

摘    要:目的:研究藿香正气液对湿阻证大鼠抗氧化作用及对胃黏膜表皮生长因子受体(EGFR)表达的影响.方法:将24只SD大鼠随机分为正常组、模型组、藿香正气液组,每组8只.正常组常规喂养,其余各组采用改进的环境加疲劳法制造湿阻证模型,连续造模6d.造模成功后,正常组和模型组予生理盐水ig(20 mL·kg-1),治疗组予藿香正气液ig (20 mL·kg-1),1次/d,连续8d.采用比色法检测血清中褪黑素(MT)、谷胱甘肽过氧化物酶(GSH-Px)及丙二醛(MDA)的含量变化,并用免疫组化方法观察胃黏膜EGFR表达的情况.结果:与正常组相比,模型组大鼠脾虚症状明显,胃黏膜有明显充血水肿,并有出血点及血块,血清MT及GSH-Px含量降低(P<0.05),MDA含量增高(P<0.05);经藿香正气液治疗后,与模型组比较,血清MT及GSH-Px含量增高(P<0.05),而MDA(P <0.05)含量降低;免疫组化结果显示模型组大鼠胃黏膜EGFR表达减少(P<0.05),藿香正气液组则明显增加(P<0.05).结论:藿香正气液可明显改善湿阻证大鼠脾虚症状并对胃黏膜损伤有保护作用,其机制可能与增强机体抗氧化应激能力及提高胃黏膜EGFR的表达量有关.

Abstract:Objective: To study the effects of Huoxiang Zhengqi liquid on the anti-oxidation and expression of the epithelial growth factor receptor(EGFR)in gastric mucosa of rats with dampness retention syndrome(DRS). Method: Twenty-four SD rats were randomly divided into 3 groups:control group,DRS group and treatment group (DRS + Huoxiang Zhengqi liquid), 8 rats in each group.DRS was induced in all rats,except the control group,by modified enviromental and fatigued method for 6 days.After DRS model was formed,control and DRS group were lavaged with normal saline(NS, 20 mL·kg-1). Rats in treatment groups were lavaged with Huoxiang Zhengqi liquid(20 mL·kg-1) for 8 days.Then the content of melatonin(MT), glutathione peroxidase(GSH-Px) and malondialdehyde(MDA) in serum were determined by colorimetric method and the expression of EGFR on gastric mucosa was determined by immunohistochemistry. Result: Rats of model groups exhibited typical DRS, with sign of hyperaemia, edema, hemorrhagic spots and sludged blood in the gastric mucosa compared to control. The content of MT,GSH-Px in serum decreased(P<0.05),while the content of MDA in serum increased(P<0.05),and the expression of EGFR in gastric musoca significantly decreased in DRS group compared to that of control group(P<0.05).Conversely, exposure of DRS rats to Huoxiang Zhengqi liquid resulted in increase in the content of MT and GSH-Px in serum(P<0.05 versus DRS group) and the expression in EGFR in the gastric mucosa(P<0.05,versus DRS group), and decrease the content of MDA in serum(P<0.05 versus DRS group). Conclusion: Huoxiang Zhengqi liquid could have the protective effect on DRS-induced gastric mucosal injury, increasing anti-oxidation and expression of EGFR may play an important role.
