中国甘肃裕固族人群6个STR位点的遗传多态性研究

目的：了解甘肃裕固族人群D3S1358、D16S539、TH01、TPOX、CSF1PO、D7S820六个STR位点遗传多态性。方法：随机抽取103名甘肃裕固族无 血缘关系个体的静脉血，应用AmpF1 STR Cofiler试剂盒扩扩增6个STR位点，PCR产物经ABI Prism377序列分析仪电泳后，用基因扫描软件进行分析。结果：6个STR位点均符合Hardy-Weinberg平衡，D2S1358、D16S539、TH01、TPOX、CSF1PO、D7S820位点的杂合度分别为0．8299，0．9384，0．8851，0．7746，0．8517，0．9436，期望排除率分别为0．4456，0．6028，0．5078，0．4267，0．5340，0．6047。个体识别率分别为0．6916，0．7820，0．7081，0．6455，0．7452，0．7863。累积DP为0．9996，累积PE为0．9886，累积PIC为0．9994。结论：裕固族群体有其自身的STR等位基因分布特征。所获得数据可为法医学个体识别、亲子鉴定及遗传学研究提供依据。

Distribution of six short tandem repeat(STR) loci in population of Yugu ethnic group in Gansu province of China

ObjectiveTo investigate the genotype and allele frequency distribution of six STR loci (D3S1358,D16S539,TH01,TPOX,CSF1PO,D7S820) in Chinese of Yugu ethnic group in Gansu province. MethodsUsing AmpFl STR Cofiler kit,six STR loci in 103 samples of Yugu ethnic group were amplified ,the PCR products were electrophresised by ABI Prism 377 sequencer,results were analyzed by Gene scan software.ResultsAll the 6 STR loci met Hardy Weinberg equilibrium.The heterozygosities (H) in D3S1358,D16S539,TH01,TPOX,CSF1PO and D7S820 were 0 829 9,0 938 4,0 885 1,0 774 6,0 851 7,0 943 6,exclusion of paternite (EPP) were 0 445 6,0 602 8,0 507 8,0 426 7,0 534 0,0 604 7,discrimination power(DP) were 0 691 6,0 782 0,0 708 1,0 645 5,0 745 2,0 786 3 respectively.The combined DP,PE and PIC were 0 999 6,0 988 6,0 999 4 respectively.ConclusionThe STR allele distribution of Yugu population has its own characteristics.The data which we obtained from Yugu population can be used not only in genetic research and population investigation,but also in human identity and paternity testing.
