副溶血性弧菌脉冲场凝胶电泳分子分型研究

摘    要:目的用脉冲场凝胶电泳方法绘制副溶血性弧菌食物中毒分离株的DNA指纹图谱,分析各菌株间的同源性关系。方法 55株食物中毒来源的副溶血性弧菌基因组DNA经限制性内切酶NotI酶切,用脉冲场凝胶电泳方法获得电泳图谱,利用BioNumerics软件对图谱进行聚类分析。结果实验得到55株副溶血性弧菌的DNA指纹图谱,可大致分为30种PFGE图谱。聚类分析显示,在大约90%的相似性水平上,有38株菌的PFGE图谱属于同一个克隆群,为2007-2008年引起闵行区食物中毒的优势流行菌型。结论闵行地区存在遗传谱系密切相关的副溶血性弧菌流行克隆。脉冲场凝胶电泳方法是分析副溶血性弧菌同源性的有效方法 ,有助于追溯传染源。

Abstract:Objective To draw a DNA fingerprint of Vibrio parahaemolyticus strains by PFGE, and to determine relationship among the Vibrio parahaemolyticus isolated from food poisoning. Methods Chromosomal DNAs from 55 strains were cut by restriction enzyme Not I, and then the molecular subtying were analyzed by PFGE. The source of these strains were analyzed using software-BioNumerics. Results There were 30 distractive PFGE patterns. 38 stains belonged to the same cluster, these strains seemed to be prevailing in Minhang in two years. Conclusions There were closely related pandemic clones of Vibrio parahaemolyticus in Minhang. PFGE was very useful method in analyzing homology of Vibrio parahaemolyticus.
