高血压大鼠肾脏ACE2的表达及其与内皮素的关系

目的探讨自发性高血压大鼠的肾脏中ACE2的表达情况并对其与血浆中内皮素水平做相关分析。方法实验分为对照组和自发高血压组，采用反转录．聚合酶链式反应（RT—PCR）法测定肾脏中ACE2mRNA表达水平，放射免疫法同批检测大鼠血浆中内皮素的含量。结果自发性高血压大鼠和正常对照大鼠相比，肾脏组织中ACE2蛋白的表达下降（P〈0．05），血清中内皮素的水平升高（P〈0．05），与ACE2蛋白的表达呈负相关（r=-0．632，P〈0．05），与血压水平呈正相关（r=0．615，P〈0．05）。结论ACE2在肾脏的表达不同可能是引发高血压病发病的一个重要因素，且其影响血压的机制可能与缩血管物质有关。

Angiotensin Converting Enzyme 2 Expression and Endothelin in Spontaneously Hypertension Rats

Objective To study the expression of angiotensin-eonverting enzyme 2 （ACE2） in spontaneously hypertension rats （SHR） and its relavanee to serum levels of endothelin（ET）. Methods Groups were divided into （1）normal：WKY rats;（2）SHR. The expression of ACE2 mRNA in kidney was determined by reverse transeription polymerase chain reaction（RT-PCR） ; Serum endothelin levels were determined by radio-immunization. Results The expression of ACF2 mRNA was decreased in kidney in SHR compared with WKY vats（ P 〈 0.05） ;Serum level of ET was increased in SHR eompared with WKY rats（ P 〈 0.05） ;The decreased expression of ACE2 was accompanied by increase of sermn ET. The level of serum ET is negatively correlated to the copy ratio of ACE2/B-aetion rnRNA in kidney（ r = - O. 632, P 〈 0.05） and positively correlated to blood pressure （ r = O. 615, P 〈 O. 05）. Conclusion The dif- ferent expression of ACE2 in kidney maybe a signifieantt factor to initiat hypertension, and the mechanism of its regulation to blond pressure is likely related to vaso-exeitor materials.
