氯胺酮抑制小鼠吗啡戒断与其抑制一氧化氮合酶的关系

摘    要:目的 观察氯胺酮抑制纳络酮引起小鼠吗啡戒断跳跃和其与一氧化氮合酶(NOS)的关系。方法 用放免法测定cAMP活性,用比色法测定NOS活性。结果 纳络酮引起吗啡依赖小鼠戒断跳跃、cAMP含量和NOS活性升高;用吗啡和氯胺酮共同处理小鼠,则显著抑制纳酮催促小鼠戒断跳跃、cAMP含量和NOS活性升高的作用。结论 氯胺酮对纳络酮引起戒断跳跃的抑制作用与其通过拮抗NMDA受体以及降低cAMP含量和NOS活性

Abstract:Objective To study the correlation between the inhibitory of naloxone-precipitated withdrawal jumps and nitric-oxide synthase (NOS) activity produced by ketamine. Methods NOS activity and cAMP content in mouse forebrain were measured by spectrophotometric analysis and radioimmunoassay respectively. Results Naloxone induced withdrawal jumps and increased NOS activities and cAMP content in the forebrain of abstinent mice. Ketamine inhibited the effects of naloxone to precipitate withdrawal jumps and increase NOS activity and cAMP content. Conclusion The inhibitory effect of ketamine on naloxone-precipitated withdrawal jumps is related to its inhibition of NOS activity and attenuation of cAMP content by blocking NMDA receptors.
