摘    要:目的:探讨乙酰胆碱受体抗体(AchRab)与重症肌无力(MG)中枢神经系统(CNS)损害之间关系。方法:将MG患者血中提取的IgG注入大量鼠尾静脉,建立大鼠被动免疫重症肌无力模型,应用免疫细胞化学技术观察Fas在脑组织中的表达,结果:模型大鼠表现出类MG症状,在MG大鼠脑内Fas抗原主要表达于大脑皮质区和海马的神经元胞膜和突起上,而在正常和空白对照组脑内没有神经细胞呈Fas抗原阳性。结论:外周注入的AchRab可能造成模型处于免疫应激状态,改变血脑屏障通透性而入脑诱发CNS损害,Fas介导的细胞凋亡可能参与MG CNS损害。

Abstract:AIM To study the relationship between Acetylcholine receptor antibody (AchRab) and dysfunction of myasthenia gravis (MG) central nervous system. METHODS The experimental MG (EMG) model was established by injection of AChRab purified from MG patient's serum into the rat's tail vein. Fas expression in brain was detected by immunocytochemistry methods. RESULTS Model rats presented the symptoms and signs which imitated MG. In EMG brains, Fas antigen was expressed on neuronal membrane anddendrite in cerebral cortex and hippocampus. However Fas antigen did not express on neuronal cell membrane and dendrite in normal and blank control group. CONCLUSION The AchRab may pass through the blood brain barrier into the brain and cause CNS dysfunction. The pathogenesis of Fas mediated apoptosis may be associated with CNS dysfunction in MG brain.
