摘    要:目的 探讨造血干细胞移植(HSCT)后外周血杀伤细胞抑制性受体P58^ 细胞比率动态变化,为防治移植物抗宿主病(GVHD)提供依据。方法 用流式细胞仪分析16例HSCT病人移植后不同时期表达P58分子的NK、CD3^+、CD6^+和CD4^+细胞的比率。结果 HSCT后外周血P58^+细胞随造血免疫功能恢复而逐渐升高,到移植后第80d时达到最高峰,随后逐渐下降。P58^ 细胞比率在CD4^ 、CD8^ 细胞的上升幅度高于NK细胞。结论 P58受异抗原激活表达,CD4^ 和CD8^ 细胞中的P58表达受抗原诱导更为明显。异基因HSC移植诱导P58表达或P58^ 细胞增殖,其生物学意义是对抗HSC移植后的过强免疫反应。

Abstract:Objective To study the varies of P58+ NK cell ratios in peripheral blood after haematopoeitic stem cell transplantation (HSCT). Hopefully this study will shield some light on the treatment of the graft verses host disease(GVHD). Method The varies of P58+ NK, CD3+, CD8+ and CD4+ cell ratios in peripheral blood after HSCT were determined by flow cytometry in 16 patients. Result P58+ cell ratios in the peripheral blood started to increase from 20 days after HSCT and peaked at 80days after HSCT and then decreased. The increase in the P58+ cell ratios of the CD4+ and CD8+ was more than that in the NK cells.Conclusion HSCT induced P58 expression or P58+ cell expansion while stimulated the immune responses. The mechanism of immunity activation stimulated P58 expression and focused on CD4+ and CD8+ subsets. Extent of P58+ cell ratios were positively correlated to GVHD development and P58+ cell ratio raised in those no GVHD patients or mild GVHD patients. The increase of P58+ ratios in CD8+ cells was higher than that of other subsets indicated that the expression of P58 in CD4+ and CD8+ cells were mainly induced by immune responses while P58 expression in NK might be constitutive.
