摘    要:目的:研究巨噬细胞移动抑制因子(MIF)、基质金属蛋白酶-9(MMP-9)和CD147在肝细胞性肝癌(HCC)组织中的表达水平及相互关系,探讨三者表达在HCC浸润、转移中的意义。方法:采用链霉亲和素-过氧化酶连接(SP)免疫组化法检测73例肝细胞性肝癌和43例肝硬化组织中MIF、CD147和MMP-9蛋白的表达。结果:①上述蛋白在HCC组织中均过表达,43例伴肝硬化的HCC病例中三者在癌组织中的表达均较肝硬化组织为高(P<0.05);②3种蛋白表达分别与肿瘤分化程度及淋巴结转移状态相关,此外MIF蛋白在有门静脉癌栓组的表达高于无门静脉癌栓组,CD147和MMP-9蛋白阳性表达率与瘤体大小相关(P<0.05);③HCC组织中MIF和CD147蛋白的表达分别与MMP-9蛋白存在显著正相关(P<0.01),MIF与CD147蛋白的表达无相关性。结论:MIF和CD147均可上调HCC组织中MMP-9的表达,三者协同表达在HCC浸润转移中起重要作用。

Abstract:Objective: To investigate the expression of macrophage migration inhibitory factor(MIF),matrix metalloproteinases(MMP)-9,and CD147,also called extracellular matrix metallo-proteinase inducer(EMMPRIN),as well as their correlation in hepatocellular carcinoma(HCC).Methods: The expression of MIF,CD147 and MMP-9 in HCC tissues of 73 cases were studied by immunohistochemical method.Results: ①All the MIF,MMP-9 and CD147 proteins were overexpressed in HCC.In the 43 cases which accompanied with liver cirrhosis(LC),expression intensities of the three proteins in cancer tissues were higher than those in LC tissues(P<0.05);②Expression of the three proteins in HCC were related to the degree of differentiation and whether lymph nodes were involved.Furthermore,the positive rate of MIF was significantly higher in carcinomas with portal vein tumor thrombus than those without,and the expression of CD147 and MMP-9 proteins were also correlated with maximal diameter of tumor(P<0.05);③The expression of MIF and CD147 proteins were positively correlated with that of MMP-9 respectively and significantly(P<0.01),but there was no relationship between the expression of MIF and CD147.Conclusion: MIF and CD147 can both up-regulate the expression of MMP-9 in HCC tissues,and the three factors have synergistic effects on the invasion and metastasis of HCC.
