胃腺癌组织MTDH的表达与临床病理特征

目的 检测和分析异黏蛋白（metadherin,MTDH）在人胃腺癌组织和癌旁正常胃黏膜组织中蛋白和mRNA表达情况,探讨MTDH的表达与临床病理特征的关系.方法 选取169例胃腺癌组织（胃腺癌组）和110例癌旁正常组织（正常组）,采用免疫组化法检测两组MTDH蛋白表达,RT-PCR法检测MTDH mRNA相对表达量,并分析MTDH蛋白、mRNA表达与胃腺癌临床病理特征的相关性.结果 胃腺癌组及正常组MTDH蛋白表达阳性率及mRNA的相对表达量分别为85.2％、22.7％和0.86±0.05、0.30±0.04,两组MTDH蛋白和mRNA表达有显著差异（P＜O.05）.不同的病理分级分期的胃腺癌MTDH蛋白和mRNA表达有显著差异（P＜0.05）,而不同的性别、年龄的胃腺癌MTDH蛋白、mRNA表达无显著差异（P＞0.05）.Sperman等级相关分析法分析显示MTDH蛋白和mRNA表达呈正相关.结论 胃腺癌组织中MTDH较癌旁正常组织中高表达;分期越高分化程度越低及发生淋巴结转移的胃腺癌组织中MTDH的表达越高.提示MTDH和胃腺癌的恶性程度和转移密切相关.

THE RELATIONSHIP BETWEEN THE EXPRESSION OF MTDH IN GASTRIC ADENOCARCINOMA TISSUES AND CLINICOPATHOLOGICAL CHARACTERISTICS

Objective To detect and analyze the MTDH in human gastric adenocarcinoma tissues and adjacent normal gastric mucosa and the expression of mRNA, to explore the relationship between MTDH expression and clini- cal pathological features. Methods We selected 169 cases of gastric adenocarcinoma （ adenocarcinoma group ） and 110 cases of adjacent normal tissue（normal group）to detect the MTDH protein expression in both groups by immu- nohistochemical method. The MTDH mRNA relative expression was detected by quantity RT - PCR method, and the correlation among the expression of mRNA, MTDH protein and clinieopathological parameters of gastric cancer were analyzed. Results Positive rates of MTDH protein expression and relative mRNA expression in gastric adenocarcino- ma group and the normal group were 85.2% ,22.7% and 0.86± 0.05,0.30 ±0.04, respectively. There was a significantly different in MTDH protein and mRNA expression between two groups （ P 〈 0.05 ）. MTDH protein and mRNA expression in different stages of pathological grading in gastric adenocarcinoma were significantly different （ P 〈 0.05 ）, but there had no significant difference concerning different gender, age of MTDH protein in gastric ade- nocarcinoma, mRNA expression （ P 〉 0.05 ）. Sperman rank correlation analysis for MTDH protein and mRNA ex- pression showed positive relation. Conclusions Expression of MTDH in gastric adenocarcinoma tissue is higher than in adjacent normal tissues;the higher staging,the less differentiation;and the higher expression of MTDH shows in gastric adenocarcinoma lymph node metastasis. It is suggested that MTDH closely related with the malignancy and metastasis of gastric adenocarcinoma.
