改良法大鼠大脑中动脉阻塞再灌注模型的制备与效果评价

摘    要:目的 通过改进大鼠大脑中动脉阻塞(MCAO)/再灌注模型的制备方法,解决模型稳定及延长存活时间问题.方法 改良方法:①将10%的水合氯醛浓度降至3.6%;②将胸骨舌骨肌—胸锁乳突肌间隙入路改为胸骨舌骨肌手术入路;③将颈外动脉(ECA)结扎或血管夹闭法改为颈内动脉(ICA)盲插结合夹镊法;④将120 min缺血再灌注并缝合皮肤时间减少至80 min.改良前、后行对照研究.结果 建模后改良组中①3.6%水合氯醛组比10%组安全范围宽,追加麻醉后死亡率低;②胸骨舌骨肌手术入路组较胸骨舌骨肌—胸锁乳突肌间隙入路组手术视野宽,血管暴露更好,出血量少,手术时间短;③调整角度后,颈内动脉(ICA)盲插结合夹镊法操作简单,较颈外动脉(ECA)结扎或血管夹闭法创伤轻,操作简便省时,且较单一盲插法成功率高;④80 min行缺血再灌注(拔除线栓)比120 min者因麻醉苏醒挣扎而产生的二次损伤少.结论 改良模型稳定,存活时间延长,能够保证神经干细胞移植方面的研究条件.

ESTABLISHMENT AND EVALUATION OF AN IMPROVED METHOD FOR RAT MIDDLE CEREBRAL ARTERY OCCLUSION AND REPERFUSION

Abstract:Objective To prepare middle cerebral artery occlusion (MCAO)and reperfusion model in rats by improving methods to establishment of a stable and long - term survival model. Methods Preparation of the model was modified as follows : 1 ) replacement of 10% chloral hydrate with 3.6% chloral hydrate ;2) replacement of the o- riginal surgical pathway between sternohyoid muscle and sternocleidomastoid muscle with per - sternohyoideus path- way ; 3 ) replacement of ligation or clipping of external carotid artery (ECA) with blind - inserting plus clipping of in-ternal carotid artery(ICA) ;4)decrease of blood flow interruption time from 120 rain to 80 min. Models prepared in old or new method were compared for evaluating the effects of modification. Results Utilization of 3.6% chloral hy- drate offered a safer space allowing the addition of anesthetics than 10% chloral hydrate. The surgical pathway by sternohyoid muscle provided wider field of vision, better blood vessel exposure, less blood loss, and shorter surgical time. Compared to ligation or clipping of ECA, blind - inserting plus clipping of ICA enjoyed advantages such as simple operation, time saving, and high success rate. Reperfusion 80 rain post blood flow interruption reduced effi- ciently the secondary injury of rats happening during revival. Conclusion The improved MCAO/reperfusion model show favorable stability and prolonging survival time, allowing the scientific researches for neural stem cell trans- plantation.
