三苯氧胺、放线菌素D和肿瘤坏死因子-a对人乳腺癌MCF-7细胞株的体外作用

摘    要:用荧光流式细胞计和细胞簇集试验证实雌激素可促使MCF-7细胞进入分裂周期,三苯氧胺作用与雌激素相反。放线菌素D在达到一定剂量时可直接杀死细胞而非抑制细胞进入分裂周期。肿瘤坏死因子-a(TNF-a)对MCF-7细胞作用类似三苯氧胺但效果远明显于三苯氧胺;放线菌素D对雌激素作用有抑制,但对三苯氧胺和TNF-a作用无拮抗。

Abstract:y means of fluorescence flow cytometer and cell cluster test,the influence of estrogen,tamoxifen,TNF-a and Actinomycin D on MCF-7 cells was determined.Results showed that estrogen enhanced MCF-7 cells into mi-tosis and tamoxifen inhibits the effect of estrogen;Actinomycin D killed MCF-7 cells rather than inhibited cell growth;TNF-a inhibited MCF-7 cells growth dramatically.The effect of estrogen on MCF-7 cell line disappeared when Actinomycin D coexisted in the medium.There was no antagonism between Actinomycin D and tamoxifen or TNF-a.
