摘    要:本文报告了紫外线照射、胰蛋白酶处理及酸抽提方法对姐妹染色单体分化染色(SCD)的影响。标本经紫外线照射(80℃30分钟)后用姬姆萨或硷性品红染色都显示正向SCD。用0.001%胰蛋白酶、pH4.0的磷酸二氢钠、20%高氯酸及5N盐酸抽提均呈反向SCD。标本经紫外线和酸双重处理,其SCD 的效应取决于先处理的手段。文章探讨了正、反向姐妹染色单体分化染色的可能机制并分析了紫外线和磷酸二氢钠处理的最佳实验条件。

Abstract:Our present study showed that sister chromatids differential staining(SCD)was influenced by various pretreatments. Chinese hamster DON cells were labeled with 13 ug/ml BudR for 29 hours and harvested by ordinary method. The slides were irradiated with UV light, stained with Giemsa or basic fuchsin. The result of SCD showed that darkly stained chromatids were located at the concave sides, while in the slides treated with 0.001% trypsin pH 4.0 NaH2PO4, 20% PCA or 5N HCl, reverse pattern of SCD was demonstrated, i. e. the darkly stained chromatids were located at the convex sides. If the slides Were treated with both UV light and acid, the pattern of cnromatids differential staining depended on the first treatment, not on the method of staining used.
