尿激酶型纤溶酶原激活物对毛囊外根鞘细胞增殖的影响

目的：探讨尿激酶型纤溶酶原激活物（Urokinase-type plasminogen activator,uPA）对体外培养的毛囊外根鞘（Outer root sheath,ORS)细胞增殖的作用。方法：体外培养了小鼠ORS细胞；用MTT法研究了uPA促进因子肝细胞生长因子（Hepatocyte growth factor,HGF)和uPA抑制因子Amiloride处理后ORS细胞地增殖变化。结果：体外培养的ORS细胞有uPA表达；HGF可促进uPA mRNA的表达并促进ORS细胞的增殖；Amiloride抑制uPa mRNA表达并抑制ORS细胞增殖。结论：毛囊ORS细胞表达uPA,HGF促进uPA的表达并促进ORS细胞增殖，Amiloride通过抑制uPA的表达和其活性抑制ORS细胞的增殖。

The effects of uPA in the proliferation of outer root sheath cells

Objective To investigate the roles of urokinase type plasminogen activator (uPA) in the proliferation of cultured outer root sheath(ORS) cells. Methods The ORS cells of the mouse vibrissa follicle were cultured in vitro . The effects of hepatocyte growth factor (HGF) and Amiloride on the proliferation of ORS cells were observed with MTT. ORS cells treated with HGF and Amiloride were analyzed by RT PCR of the uPA gene. Results The ORS cells expressed uPA mRNA; HGF promoted and Amiloride inhibited the uPA mRNA production and the proliferation of ORS cells. Conclusion ORS cells could express uPA mRNA, HGF might promote the expression of uPA and the proliferation of ORS cells through inhibiting the expression and the activation of uPA, Amiloride inhibit the proliferation of ORS cells.
