环氧合酶-2在子宫肌瘤组织中的表达及其意义

摘    要:目的观察环氧合酶-2(cyclooxygenase,COX-2)在子宫肌瘤与子宫平滑肌组织中的表达,探讨COX-2与子宫肌瘤发病机制的相关性。方法选择2010年1月至2010年8月在上海市杨浦区中心医院行腹腔镜下子宫肌瘤切除术的30例患者的子宫肌瘤组织为实验组,并取其邻近的正常平滑肌组织为对照组,采用免疫组化方法检测COX-2的表达;并采用蛋白印迹法及实时荧光定量逆转录聚合酶链反应分别在蛋白水平和基因转录水平检测标本中COX-2的表达。结果免疫组化检测实验组和对照组均有COX-2蛋白表达,平滑肌细胞阳性指数(MPI)为11.90,子宫肌瘤细胞阳性指数(FPI)为46.50,两者比较,差异有统计学意义(P〈O.05)。蛋白印迹法结果显示,COX-2在实验组(O.872±0.035)中的表达明显高于对照组(O.202土0.056),两者比较,差异有统计学意义(P〈0.05)。COX-2mRNA在实验组(0.122±0.062)中的表达也高于对照组(0.025±0.009),两者比较,差异有统计学意义(P〈0.05)。结论COX-2在子宫肌瘤的表达明显高于子宫平滑肌,COX-2可能在子宫肌瘤的发病机制中起重要作用。

Abstract:Objective To detect the expression of cyclooxygenase - 2 (COX- 2) in uterine fibroids and normal uterine smooth muscles thus to investigate its roles in the pathogenesis of uterine fibroids. Methods To detect the expression of cyclooxygenase - 2 in uterine fibroids and normal uterine smooth muscle thus to investigate its roles in the pathogenesis of uterine fibroids. Results the COX - 2 positive index in smooth muscle cells (11.90) was much lower than that in uterine fibroids cells (46.50, P〈0.05). At protein level, the COX- 2 expression in uterine fibroids was significantly higher than in smooth muscle (0. 872 4±0. 035 vs 0. 202 ±0. 056). Similarly, at mR- NA level, COX- 2 expression in uterine fibriods (0. 122 ± 0. 062) was higher than in normal smooth muscle tissue (0. 025 4± 0. 009) (P〈0. 05). Conclusion COX- 2 expression in uterine fibroids was significantly higher than uterine smooth muscles, suggesting an imoortant role nf COX - 2 in the pathogenesis of uterine fibroids.
