血清HER2检测在乳腺肿块中的应用

目的 建立血清HER2水平检测方法,研究乳腺肿瘤患者血清HER2的水平、影响因素及临床意义,为乳腺癌治疗及预后判断提供参考.方法 采用酶联免疫吸附(ELISA)方法检测10例正常人、20例乳腺良性病变、50例早期手术及13例晚期转移性乳腺癌患者血清HER2的水平,并对乳癌手术组患者用免疫组化检测组织HER2/neu的表达情况.结果 健康对照组、乳腺良性病变组及乳癌手术组之间的血清HER2水平及阳性率无明显差异(P＞0.05),晚期转移性乳腺癌组的血清HER2水平及阳性率与前三者之间有显著性差异(P＜0.05);乳癌手术组血清HER2水平与肿瘤大小有相关性(P＜0.05),与组织HER2/neu表达、淋巴结转移无关(P＞0.05).结论 血清HER2的水平与乳腺肿块的肿瘤负荷密切有关,可作为免疫组化的补充,用于监测复发转移性乳腺癌并指导其治疗.

Clinical Application of Serum HER2 in Patients with Breast Cancer

Objective To establish a method to detect the level of serum HER2 in patients with breast cancer and study relationship between level and tissue HER2/neu overexpression and other factors. Methods A total of 83 females were included in the study including 10 normal controls,20 benign breast disease patients,50 primary breast cancer and 13 metastatic breast cancer patients.The level of serum HER2 was detected with method of ELISA.The HER2/neu overexpression of the primary tumor of breast cancer was measured by the IHC.Results We observed the positive rate and level of HER2 in sera of healthy subjects and benign breast disease patients was no difference to those of operable breast cancer patients(P>0.05),but significant difference to those of metastatic breast cancer patients(P<0.05).There was a correlation between serum HER2 and tumor size(P<0.05),there was no correlation between serum HER2 and axillary lymph node or tissue HER2/neu overexpression(P>0.05).Conclusion There is correlation between serum HER2 and tumor loads of breast cancer.It could be helpful in monitoring recurrent or metastatic breast cancer and directing treatment.
