瘦素对多囊卵巢综合征患者卵巢黄素化颗粒细胞雌激素分泌的影响

摘    要:目的 探讨不同浓度瘦素在促卵泡激素(FSH)的参与下对多囊卵巢综合征(PCOS)患者卵巢黄素化颗粒细胞雌二醇(E2)的影响.方法 20例行体外受精-胚胎移植(IVF-ET)的不孕症患者,分成PCOS组和对照组各10例.采用卵泡刺激长方案促排卵,体外培养得黄素化颗粒细胞,然后在不同浓度瘦素(0 ng/ml、10 ng/ml、20 ng/ml、100 ng/ml、1 000 ng/ml)、卵泡刺激素(FSH)(5 mIU/ml)及雄烯二酮(1 μmol/ml)联合作用下继续进行培养24 h,收集培养液,用放射免疫法测定颗粒细胞产生E2的浓度.结果 加入不同浓度瘦素(10 ng/ml、20 ng/ml、100 ng/ml、1 000 ng/ml)后PCOS组和对照组卵巢颗粒细胞E2的分泌均受到明显抑制;随瘦素浓度的增加E2水平逐渐降低(P<0.05).结论 瘦素能抑制PCOS卵巢黄素化颗粒细胞雌激素的分泌,其抑制作用随瘦素浓度的增加而增强.

Abstract:Objective To investigate the effects of leptin on estradiol secretion of human luteinized cells of polycystic ovarian syndrome (PCOS)in vitro in combined with follicle stimulating hormone (FSH). Methods osa All patients were divided into two groups, who were given follicle-stimulating therapy for ovulation induction, human luteinized granulosa cells of the two groups, from in-vitro fertilization-embryo transfer(IVF-ET) : PCOS atom) and normal control group,which were cultured in vitro after purification, and then cultured with medium plus various concentration of letpin(0 ng/ml, 10 ng/ml, 20 ng/ml, 100 ng/ml, 1 000 ng/ml ) and FSH (5 mIU/ml ) combined with androstenedione (1 μmol/ml)for 24 hours. The media was collected to measure the concentration of estrogen production by luteinized granulosa cells. Results Different concentrations of leptin ( 10 ng/ml, 20 ng/ml, 100 ng/ml, 1 000 ng/ml) inhibited PSH-stimulated estradiol production by ovarian luteinized granulosa cells in PCOS group and normal group. The production of estradiol decreased with the concentration of leptln increase(P 〈0. 05). Conclusion Leptin cause a concentration-related inhibition of FSH-stimulated estradiol production in luteinized granulosa cells of PCOS, the inhibiting effect increases along with the elevation in leptin concentration.
