蒙药平喘灵胶囊防治支气管哮喘过程中的相关因子分析研究

摘    要:目的:探讨蒙药平喘灵胶囊对哮喘模型小鼠气道炎症及白细胞介素17A(IL-17A)、转化生长因子-β1(TGF-β1)、肿瘤坏死因子-α(TNF-α)、Smad2/3和Smad7的影响。方法:清洁级雌性BALB/c小鼠60只,随机分为正常对照组、模型对照组、地塞米松片组及平喘灵胶囊1、2、4 g/kg组,采用鸡蛋清白蛋白(OVA)建立实验性哮喘小鼠模型,激发第21 d开始灌胃给予相应药物,模型对照组和正常对照组以生理盐水代替,连续给药8 w。末次给药后24 h内,眼球取血处死,细胞计数器计数肺泡灌洗液(BALF)中白细胞分类数目;HE染色观察小鼠肺组织病理变化;免疫组织化学法(IHC)检测肺组织Smad2/3、Smad7蛋白相对表达水平;酶联免疫吸附试验(ELISA)检测小鼠BALF及血清IL-17A、TGF-β1、TNF-α的含量。结果:与正常对照组比较,模型对照组BALF白细胞分类、BALF及血清IL-17A、TNF-α及TGF-β1含量显著升高、哮喘小鼠肺组织smad2/3蛋白表达水平显著下调(P<0.01);小鼠肺组织Smad7蛋白表达水平显著下调(P<0.01)。与模型对照组比较,蒙药平喘灵胶囊各组BALF白细胞分类、BALF及血清中IL-17A、TNF-α及TGF-β1含量显著降低、哮喘小鼠肺组织Smad2/3蛋白表达水平显著下调,小鼠肺组织Smad7蛋白表达水平显著上调(P<0.01)。结论:蒙药平喘灵胶囊对哮喘小鼠气道炎症有较好的改善作用,其机制可能与降低BALF及血清IL-17A、TNF-α、TGF-β1含量及下调小鼠肺组织smad2/3蛋白表达水平和上调小鼠肺组织smad7蛋白表达水平有关。

Correlation Factors of Mongolia Medicine Pingchuanling Capsule in Preventing and Treating Bronchial Asthma

Abstract:Objective:To investigate the effects of Mongolian medicine Pingchuanling capsule on airway inflammation,interleukin 17 A(IL-17 A)、transforming growth factor-β1(TGF-β1)、tumor necrosis factor-α(TNF-α),smad2/3 and smad7 in asthma model mice.Methods:60 female BALB/c mice were randomly divided into the model group,the control group、Dexamethasone group、Pingchuanling capsule groups(1、2、4 g/kg).OVA(egg white protein)was used to establish the experimental asthma mice model.On the 21 st day of the stimulation,mice were orally administered with corresponding drugs or saline water,for 8 weeks.Within 24 hours after the last administration,blood was taken from the eye ball and mice were executed.The classification numbers of white blood cells in BALF were counted by cell counter.HE staining was used to observe the pathological changes of lung tissues.The relative expression levels of smad2/3 and smad7 proteins in lung tissues were detected by immunohistochemistry(IHC).The contents of IL-17 A,TGF-β1 and TNF-αin BALF and serum of mice were determined by enzyme-linked immunosorbent assay(ELISA).Results:Compared with the control group,BALF white blood cell classification,IL-17 A、TNF-αand TGF-β1 in BALF and serum,and smad2/3 in lung tissues of asthmatic mice were significantly increased in the model group(p<0.01).The expression level of smad7 was significantly down regulated(p<0.01).Compared with the model group,the BALF white blood cell classification,IL-17 A、TNF-αand TGF-β1 in BALF and serum,the smad2/3 in lung tissues of asthmatic mice were significantly reduced in Mongolian medicine Pingchuanling capsule group.The expression of smad7 in the lung tissue was significantly up regulated(p<0.01).Conclusion:Mongolian medicine Pingchuanling capsule shows the good effect on airway inflammation in asthmatic mice,its mechanism may be related to down-regulating the contents of IL-17 A、TGF-β1 and TNF-αin BALF and serum,reducing the expression level of smad2/3 and up-regulating the smad7 expression level in the lung tissue.
