黑米花色苷对白血病细胞株HL-60及正常淋巴细胞活性氧及线粒体膜电位的影响

摘    要:目的 观察黑米花色苷对白血病HL-60细胞和正常淋巴细胞活性氧(reactive oxygen species,ROS)水平及线粒体膜电位的影响.方法 以2,7-二氯荧光素(2,7-dichloro fluoreseeineiactate,DCFH-DA)为细胞内活性氧探针,以罗丹明-123为反映线粒体膜电位荧光探针,流式细胞仪和激光共聚焦扫描显微镜检测分析黑米花色苷作用下HL-60细胞及正常淋巴细胞活性氧水平及线粒体膜电位的变化.结果 黑米花色苷处理正常淋巴细胞2、24 h其ROS水平未见显著变化(P>0.05);黑米花色苷处理HL-60细胞,ROS水平首先显著升高(P<0.01),随后逐渐降低,并在12 h显著低于对照纽(P<0.01).黑米花色苷处理正常淋巴细胞24 h线粒体膜电位无显著变化(P>0.05),处理HL-60细胞24 h线粒体膜电位显著降低(P<0.01).结论 黑米花色苷能够显著影响HL-60细胞活性氧水平,并显著降低HL-60细胞线粒体膜电位,而对正常淋巴细胞活性氧及线粒体膜电位无显著影响.
