摘    要:目的建立人胰液蛋白表达谱。方法采用丙酮沉淀法处理来自于四个非胰腺癌患者的胰液标本,利用双向凝胶电泳和基质辅助激光解析电离飞行时间串联质谱(MALDI-TOF-MS/MS)技术对胰液蛋白进行分离和鉴定。结果建立了高分辨率的人胰液蛋白表达谱,鉴定了91个独立的胰液蛋白,并对91个蛋白进行了功能注释分析。结论胰液蛋白表达谱为深入理解胰腺疾病的病理生理机制及筛选和发掘胰腺癌潜在生物标志物提供了理论基础。

Abstract:Objective To construct an expression profiling of human pancreatic juice protein.Methods Pancreatic juice samples from four patients who were not suffering from cancer of the pancreas or ducts were treated by using an acetone precipitation method.Pancreatic juice proteins were separated and identified by using two-dimensional electrophoresis(2-DE) and matrix-assisted laser desorption/ionization-time of flight tandem mass spectrometry(MALDI-TOF-MS/MS).Results The expression profiling with a high resolution of human pancreatic juice proteins was constructed.Ninety-one unique proteins were identified and functional annotation analysis was performed.Conclusions Protein expression profiling can provide a theoretical basis for further understanding the physiological and pathological mechanisms involved in pancreatic diseases,and screening and for finding potential biomarkers for pancreatic cancer.
