左旋多巴对健康老年大鼠的神经毒性

摘    要:目的研究长期应用左旋多巴对健康老年大鼠多巴胺神经元是否存在毒性作用。方法将24只健康老年大鼠随机分为3组,分别给予大剂量左旋多巴、小剂量左旋多巴、生理盐水共4个月,观察大鼠用药后是否产生运动障碍,以及纹状体部位脑组织匀浆中的还原型谷胱甘肽、谷胱甘肽过氧化物酶、超氧化物歧化酶、丙二醛含量,以免疫组化法检查中脑黑质和纹状体部位酪氨酸羟化酶阳性细胞和纤维的数量。结果没有发现长期应用左旋多巴会引起运动障碍,长期左旋多巴应用引起纹状体部位还原型谷胱甘肽、谷胱甘肽过氧化物酶含量增加,超氧化物歧化酶、丙二醛含量无明显变化;黑质和纹状体部位酪氨酸羟化酶阳性细胞数在各组间无明显差别。结论长期应用左旋多巴对健康老年大鼠无神经毒性作用。

Abstract:Objective To identify the neurotoxicity of chronic levodopa in healthy aged S-D rats.Methods 24 healthy aged S-D rats were divided into three groups,which was respectively gavaged with high-dose of L-dopa(150 mg/kg),low-dose of L-dopa(50 mg/kg),0.9%NS for 4 months.We observed the rats of involuntary abnormal mobility before and after levodopa.After chronic levodopa for 4 months,we measured the oxidive stress with glutathione,glutathione peroxidases,superoside dismutase,maleic dialdehyde.Immunohistochemistry was used to observe the numbers of TH-stained neurons in the substantia nigra and fibers in striatum.Results IAM was not found,glutathione and glutathione peroxidases in the striatum slices were increased.Superoxide dismutase and maleic dialdehyde were unchanged.There was no difference in the numbers of TH-stained neurons and fibers in striatum among three groups.Conclusion The chronic levodopa has no neurotoxicity in healthy aged S-D rats.
