消积饮影响小鼠Lewis肺癌细胞增殖与凋亡的原位研究

摘    要:目的:研究消积饮对小鼠lewis肺癌的抑制作用及其分子机制。方法:复制小鼠lewis肺癌模型,观测消积饮对其瘤体大小的影响,并与替加氟组、生理盐水组等进行比较;同时,运用免疫组织化学及DNA片段末端标记法检测PCNA的表达和凋亡细胞的数目在各实验组用药前后的变化。结果:消积饮组和替加氟组平均瘤重、PCNA表达阳性率均低于生理盐水组(P<0.01),凋亡细胞数目高于生理盐水组(P<0.01);而上述二项指标的改变在消积饮组和替加氟组间则无显著性差异(P>0.05)。结论:消积饮对小鼠lewis肺癌细胞确有抑制效应,其机理可能与消积饮抑制细胞增殖和诱导细胞凋亡有关。

Abstract:Objective:To study the inhibition and its molecular mechanism of Xiaoji Decoction on lewis lung carcinoma in mice Methods:Lewis lung carcinoma model of mice was duplicated.The effect of Xiaoji decoction on the size of tumor,the expression of proliferative cell nucleus antigen(PCNA)and the number of apoptosis cells were observed,and compared with those of the Tagafer group and the control group by immunohistochemical methods and using in situ end labeling technique.Results:by comparison with the control group,the mean tumor size and the expression of PCNA in Xiaoji decoction group and in the Tegafur group were reduced ( P <0 01),and the number of apoptosis cells increased( P <0 01);There were no significant statistic differences between the mean tumor size,the expression of PCNA and the apoptosis number.Conclusion:Xiaoji decoction has definite effect in treating lewis lung carcinoma of mice.The mechanism may be through inhibiting the expression of PCNA and inducing apoptosis.
