| 摘 要： | 目的观察不同浓度的过氧化氢(H2O2)对培养的大鼠主动脉血管平滑肌细胞(VSMC)骨桥蛋白(OPN)基因表达的影响。方法用含10%小牛血清的DMEM培养液体外培养大鼠主动脉VSMC,随机分为对照组,H2O2不同浓度(10,50,100μmol/L)干预组,应用逆转录聚合酶链反应及Western blot技术结合吸光度扫描分析,观察H2O2对VSMC的OPN表达的影响。结果不同浓度H2O2均明显刺激VSMC的OPN mRNA和蛋白的表达,且具有剂量依赖性促进作用。结论H2O2能在基因水平上刺激VSMC的OPN的表达。

| Abstract: | Objective To investigate the effect of H_2O_2 on osteopontin gene expression in rat aortic vascular smooth muscle cell(VSMC).Methods Rat aortic vascular smooth muscle cells were cultured in vitro in DMEM containing 10 percent calf serum.The cells were incubated with different concentrations of H_2O_2,then RT-PCR and Western bloting analysis were used to observe the effect of H_2O_2 on osteopontin expression in rat VSMC.Results Different concentrations of H_2O_2 could obviously induce gene expression and protein synthesis of osteopontin,and exhibited dose-dependent promotion effect.Conclusion The results demonstrated that H_2O_2 could stimulate osteopontin gene expression in rat VSMC. |
