马来酸曲美布汀对幼鼠肠易激综合征模型肠黏膜的保护作用

摘    要:目的 探讨马来酸曲美布汀治疗幼鼠肠易激综合征(IBS)模型腹壁肌电活动及肠黏膜上皮细胞功能的影响.方法 建立幼鼠IBS模型16只,随机选8只用马来酸曲美布汀治疗为治疗组,另8只为阳性对照组,另取同期正常生长幼鼠8只为阴性对照组.分别对3组幼鼠行肛门直肠球囊扩张刺激下测腹壁肌电活动;处死后取结肠对其肠黏膜上皮细胞进行分层刮片、荧光染色和还原能力测定.结果 与阴性对照组比较,阳性对照组腹壁肌电活动随扩张压力增高而明显增强(P<0.05),治疗组与阴性对照组之间差异无统计学意义(P>0.05).幼鼠IBS模型肠黏膜上皮细胞功能处于抑制状态,马来酸曲美布汀治疗能改善这种抑制.结论 马来酸曲美布汀能改善幼鼠IBS肠黏膜上皮细胞功能,抑制其内脏高敏感性.

Abstract:Objective To investigate the effect of trimebutine maleate on the changes of abdominal wall myoelectric activity and intestinal epithelial cells function in the young rat model with irritable bowel syndrome(IBS).Methods Sixteen young rats IBS models were established and randomly divided into the treatment group with trimebutine maleate(T) and positive control group(PC) with 8 rats each.At the same time,another 8 infant healthy rats were chosen as the negative control group(NC).Abdominal wall myoelectric activities were measured when the anus and rectum were distented by sacculus vestibularis in the three groups.The intestinal epithelial cells were sliced and stained fluorescently.The reduction ability was determined.Results The abdominal wall myoelectric activity of group PC was obviously greater than that of group NC with the increased distention pressure(P<0.05),which was not significantly different between the group T and group NC(P>0.05).The function of the intestinal epithelial cells in IBS young rats was in the inhibitory state,which was improved after treated with trimebutine maleate.Conclusion Trimebutine maleate can inhibit the visceral hypersensitivity and improve the function of the intestinal epithelial cells in IBS young rats.
