川芎嗪、山莨菪碱、蝮蛇抗栓酶及卡托普利对实验性糖尿病大鼠肾脏的保护作用

摘    要:目的 探讨川芎嗪、山莨菪碱、蝮蛇抗栓酶及卡托普利对实验大鼠糖尿病肾病(DN)的预防作用。方法 SD大鼠60只,分为正常对照组、糖尿病对照组及各糖尿病治疗组。以四氧嘧啶腹腔内注射制成糖尿病模型。各治疗组分别给予上述药物治疗,每周5次,共14周。取肾脏行光镜和电镜检查,并进行图像分析,同时行蛋白糖基化产物测定。结果 与糖尿病对照组比较,各治疗组大鼠肾组织中蛋白糖基化产物水平明显降低(P<0.01);除蝮蛇抗栓酶组外,其他治疗组大鼠肾小球截面积亦明显缩小(与糖尿病对照组比较,P<0.05);电镜显示,糖尿病对照组大鼠肾小球基底膜显著的不均匀增厚,而各治疗组病变较轻。上述各种药物疗效均以卡托普利组最为明显。结论 改善微循环、血液流变学及 血流动力学可减慢实验大鼠DN的进展。

Abstract:Objective To study the prophylactic effect of tetramethylpyrazine, anidamine, ahalysan tinfarctase and captopril for diabetic nephropathy (DN). Methods 60 SD rats were divided randomly into normal control group(NC), diabetic control group (DC) and diabetic treatment group (DT). Diabetes model was established by intraperitoneal injection of alloxan. T etramethylpyrazine, anidamine, ahalysan tinfarctase and captopril were given respectively to each DT group 5 times a week and for 14 weeks. The kidneys were processed for morphological examination with light and electron microscopes, and the glucosylation of protein (GP) in renal tissue was assayed. Results Compared with DC, the level of GP in renal tissue in each DT group was decreased significantly (P<0.01), and the glonerular plana atea in each DT group eas also reduced (P<0.05) except for the ahalysan tinfarctase-treated group. Electron microscopic findings indicated that the glomerular basement membrane in DC rats was unevenly thick, and these abnormal changes were alleviated significantly in each DT rat especially in the captopril-treated group. Conclusion Improving microcirculation, blood rheology and hemodynamics could slow the progress of the DN in the experimental diabetes rats.
