| 摘 要： | 目的:观察三七总皂苷含药血清对溶血磷脂酸(LPA)诱导的血管平滑肌细胞增殖效应的影响,分析该作用与细胞外信号调节激酶ERK信号通路的关系。方法:采用贴块法培养大鼠主动脉平滑肌细胞;以四甲基偶氮唑蓝(MTT)比色法观察不同分组对细胞增殖的影响;采用流式细胞仪分析细胞周期和DNA含量。结果:空白血清+LPA组较空白血清组OD值升高(P<0.05),S期指数升高(P<0.01);三七总皂苷含药血清+LPA组与卡托普利含药血清+LPA组比较OD值降低(P<0.05),S期指数无差异;三七总皂苷含药血清+LPA+PD98059组较空白血清组OD值降低(P<0.05),S期指数无差异。结论:三七总皂苷对LPA诱导的VSMC增殖有拮抗作用,且作用强于卡托普利,其作用机制可能与抑制LPA对ERK信号通路的激活,从而抑制DNA合成,阻止细胞进入S期有关。
