过敏性紫癜急性期患儿淋巴细胞活化、凋亡及雷公藤对其作用

摘    要:目的探讨急性期过敏性紫癜(HSP)患儿外周血淋巴细胞(PBLs)活化、凋亡及雷公藤内酯醇(TP)对其作用。方法选择PBLs为研究标的细胞,以流式细胞仪检测CD3、CD25抗原表达及凋亡率。结果与健康儿童比较,HSP患儿PBLs培养0、48 h后CD3 、CD25 细胞百分率均明显升高(P<0.05),凋亡率均明显降低(P<0.05);应用TP后培养48 h HSP患儿PBLs CD3 CD25 细胞百分率明显降低(P<0.05),凋亡率明显升高(P<0.05)。结论HSP患儿PBLs活化明显增多,凋亡延迟,可能参与HSP的发病;TP抑制PBLs活化并诱导其凋亡,可能是雷公藤对HSP作用机制之一。

Abstract:Objective To explore the activation and apoptosis of peripheral blood lymphocytes(PBLs) in children with Henoch-Schonlein purpura(HSP) and the effects of triptoIide(TP) on them. Methods The changes of activation and apoptosis were observed on cultured PBLs in children with HSP and healthy controls ,and the effects of TP were compared respectively. Expression of CD3, CD25 and apoptosis rate of PBLs were assayed with flow cytometry. Results The percentage of CD3 CD25 cell was significantly higher (P<0.05)and the percentage of apoptosis was significantly lower(P<0.05)in PBLs cultured for zero hour and 48 hours in children with HSP than that in healthy children respectively. After treated with TP, the percentage of CD3 CD25 cells decreased significantly (P<0.05)and the percentage of apoptosis increased significantly(P<0.05)in PBLs in children with HSP.Conclusions There exist multiple immune regulatory disturbances in HSP patients, the activation of PBLs is raised, apoptosis is delayed significantly. TP has obvious immune regulatory effects, which can inhibit activation of T cells and raise apoptosis.
