PTEN反义寡核苷酸对辛伐他汀调控心肌细胞肥大的影响

目的:观察抑癌基因—PTEN的反义寡核苷酸对辛伐他汀调控心肌细胞肥大的影响。方法:应用图像分析系统及[3H]-亮氨酸掺入法分别测定心肌细胞表面积及蛋白合成速率,RT-PCR检测心钠素(ANP)mRNA表达,RT-PCR和West-ern blot法检测PTEN的mRNA和蛋白表达水平。结果:辛伐他汀 PTEN反义链组心肌细胞表面积、[3H]-亮氨酸掺入率、ANP mRNA水平均明显高于辛伐他汀组(P<0.01),PTEN mRNA及蛋白表达水平明显低于辛伐他汀组(P<0.01)。结论:PTEN反义寡核苷酸能够抑制PTEN表达、部分逆转辛伐他汀对心肌细胞肥大的抑制作用。

Modulation effects of PTEN antisense oligodeoxynucleotides on the inhibition of rat cardiac myocytes hypertrophy by simvastatin

Objective:To study the modulation effects of PTEN antisense oligodeoxynucleotides on the inhibition of cultured rat cardiac myocytes hypertrophy by simvastatin.Methods: Image analysis system was used to measure cell surface area.Protein synthesis was measured via -leucine incorporation.The mRNA level of atrial natriuretic peptide(ANP) was determined with RT-PCR.To study the mRNA and protein level of PTEN,RT-PCR and Western blot were applied,respectively.Results: Cell surface area,-leucine incorporation rate,ANP mRNA levels in simvastatin PTEN antisense oligodeoxynucleotides group were significantly higher than those of simvastatin group(P<0.01).The mRNA and protein levels of PTEN in antisense oligodeoxynucleotides group were significantly lower than those of simvastatin group(P<0.01).Conclusion: PTEN antisense oligodeoxynucleotides can inhibit PTEN expression,thus reverse the inhibition effects of simvastatin on cardiac myocytes hypertrophy to some extend.
