异丙酚预处理对缺血再灌注脑损伤大鼠中性粒细胞弹性蛋白酶的影响

摘    要:目的探讨异丙酚预处理对大鼠脑缺血再灌注(I/R)损伤脑组织中性粒细胞弹性蛋白酶(NE)的影响。方法SD大鼠30只,随机分为正常对照组(C组),脑缺血再灌注组(I/R组),异丙酚预处理组(P组,于脑缺血前10min经腹腔注射异丙酚100mg/Kg)(n=10)。采用大鼠大脑中动脉线栓法建立局灶性脑I/R损伤模型。采用ELISA法检测脑组织NE、TNF-α、IL-1β的表达。结果与C组比较,I/R组和P组NE的活性及TNF-α、IL-1β的含量均明显升高(P〈0.01);与I/R组比较,P组NE的活性及TNF-α、IL-1β的含量均明显降低(P〈0.01)。结论异丙酚预先给药能够通过抑制脑I/R后脑组织内NE的活化进而抑制TNF-α、IL-1β的过度表达,降低炎性反应,对脑I/R损伤起到保护作用。

Abstract:Objective: To investigate the effect of propofol pretreatment on the expression of neutropil elastase(NE) following cerebral ischemia-reperfusion(IR) injury in rats.Methods:Thirty healthy male SD rats,weighing 240~280 g,were randomly divided into the control group(Group C),cerebral ischemia-reperfusion group(Group IR) and propofol pretreatment group(Group P,propofol was injected intraperitoneal 10 min before cerebral ischemia)(n=10).Focal cerebral ischemia-reperfusion model was established in rats by reversible middle cerebral artery occlusion with filament.Cerebral ischemia was for 2 hours and then with 24 hours of reperfusion.Neurological deficit score was executed after cerebral ischemia 2 h and reperfusion 24 h in the rats.ELISA assay was used to detect the expression of NE,TNF-α and IL-1β in the brain tissue.Results:Compared with Group C,the activity of NE and the expression of TNF-α and IL-1β in Group IR and Group Propofol were significantly increased(P<0.01);Compared with Group IR,the activity of NE and the expression of TNF-α AND IL-1β in Group Propofol were significantly decreased(P<0.01).Conclusion:Different dose propofol pretreatment can significantly reduce the activation of NE and further,inhibit the over-expression of TNF-α and IL-1β in the brain tissue following cerebral IR injury,and reduce the inflammatory response,and therefore has a significantly protective effect on cerebral IR injury.
