摘    要:目的初步探讨活血汤在脑缺血再灌注损伤过程中的神经保护作用及机理。方法采用大脑中动脉阻断法(middle cerebral antety occlusion,MCAO)制备大鼠脑缺血再灌注模型,随机分组。观察药物对大鼠的神经功能缺损症状、脑梗死体积、单位神经元密度的影响;测定脑组织一氧化氮合酶(NOS)、丙二醛(MDA)、超氧化物歧化酶(SOD)含量;免疫印迹分析 NMDA 受体亚单位蛋白(NR1)的表达。结果活血汤能改善脑缺血再灌注后的神经功能缺损症状,减小梗死体积,减少神经元损伤;活血汤能显著降低脑组织内NOS 活性,NR1表达,减少 MDA 含量和增加 SOD 活性。结论活血汤对大鼠脑缺血再灌注损伤具有神经保护作用,为进一步临床应用提供实验理论依据。

Protective Effect of Huoxue Decoction on Cerebral lschemia/Reperfusion Injury in Rats

Abstract:Objective To preliminarily explore the nerve protective function and the mechanism of Huoxue Decoction (HXD) in the process of cerebral ischemia/reperfusion injury in rats.Methods Cerebral ischemia/reperfusion rat model was es- tablished by adopting middle cerebral artery occlusion (MCAO) and randomly grouped.Effects of HXD on nerve function deficit syndrome,on the volume of cerebral infarction,and on per unit neuron density were observed.Contents of nitric ox- ide synthase(NOS),malondialdehyde(MDA),superoxide dismutase(SOD) in brain issue were determined.Expression of NMDA receptor subunit protein NR1 was determined by immunoblotting technique.Results HXD could improve the nerve function deficit syndrome after cerebral ischemia/reperfusion,lower the volume of cerebral infarction,and lower the injury of neurons.Besides,HXD could significantly lower the NOS activity of cerebral tissues and NR1 expression,lower MDA content and increase SOD activity.Conclusion HXD had nerve protective function of cerebral ischemia/reperfu- sion injury in rats,which further provided an experimental theoretical basis for clinical application.
