川芎嗪抑制大鼠颈动脉球囊损伤后再狭窄的在体研究

摘    要:研究川芎嗪对大鼠颈动脉球囊损伤后再狭窄的影响及机制。方法建立SD大鼠颈动脉球囊损伤模型,分为假手术组、模型组和川芎嗪组。川芎嗪组为川芎嗪10mg(kg·d)]尾静脉预给药1周,球囊成形术后继续同剂量川芎嗪尾静脉注射6周。于第6周末收集损伤段颈动脉,病理切片HE染色测量内中膜厚度比及Western blot检测相关蛋白表达水平。结果大鼠颈动脉球囊损伤后6周,损伤段血管内膜明显增厚,模型组和川芎嗪组与假手术组比较,内膜/中膜面积比显著增加(P<0.05);与模型组比较,川芎嗪组内膜/中膜面积比下降(P<0.05)。模型组损伤段血管Wnt 4、Dvl-1、β- catenin、Cyclin- D1表达表达水平较假手术组显著增高,而川芎嗪组的表达明显减少(均P<0.05)。结论川芎嗪可以抑制大鼠颈动脉球囊损伤后再狭窄,其机制与下调Wnt信号通路有关。

Tetramethylpyrazine prevents balloon-injured carotid artery from restenosis in rats

Abstract:Objective To investigate the preventive effect of tetramethylpyrazine (TMP) on bal oon- injured rat carotid artery from restenosis. Methods Bal oon- injured carotid artery model was established in SD rats. SD rats were divided into 3 groups:sham operation group(n=8);model group(n=8) and TMP group(n=8). In TMP group, rats were pretreated with caudal vein injection of TMP (10mg/kg·d) for 1 week, and the same dose of TMP was continued for 6 weeks after bal oon angioplasty. Injured segments of carotid artery were harvested at 6th week and embedded in paraffin wax to assess the size of intimal lesions by HE staining. Proteins expressions of Wnt4, Dvl- 1,β- catenin and Cyclin- D1 were detected by Western blotting method. Results Six weeks after bal oon injured, the intimal of injured artery became thickening. The ratio of intima/tunica media thickness and col-lagen volume in intima increased significantly compared to those of sham operation group (P〈0.05). Compared to model group, the ratio of intima/tunica media thickness in TMP group reduced significantly (P〈0.05). The expressions of Wnt4, Dvl- 1,β- catenin and Cyclin- D1 proteins in model group increased significantly in comparison with sham operation group, while de-creased in comparison with TMP group. Conclusion TMP can inhibit restenosis of bal oon- injured carotid artery possibly through down- regulation of wnt signal pathway.
