E- cadherin、CD44v6、MMP2在膀胱尿路上皮癌中的表达及意义

摘    要:目的:研究钙黏附分子E- cadherin(E- cad)、CD44变异体6(CD44v6)和基质金属蛋白酶(MMP2)在膀胱尿路上皮癌(bladder urothelial carcinoma)中的表达及对预后的意义。方法应用免疫组化Envision法,检测105例膀胱尿路上皮癌患者和20例正常体检者膀胱黏膜E- cad、CD44v6及MMP2的表达情况。结果20例正常膀胱黏膜中E- cad、CD44v6均正常表达,MMP2均阴性表达,105例膀胱尿路上皮癌中E- cad及CD44v6的异常表达率分别为43.8%(46/105)和58.1%(61/105),MMP2阳性表达为53.3%(56/105)。E- cad、CD44v6的异常表达率和MMP2的阳性表达率与患者肿瘤的病理分级、临床分期及生存率相关。结论E- cad、CD44v6的异常表达和MMP2的阳性表达在膀胱尿路上皮癌的演变过程中起着重要作用,可作为监测肿瘤恶性程度和预后有效的分子生物学指标之一。

Expression of E-cadherin,CD44v6 and matrix metalloproteinase-2 in bladder urothelial carcinoma and its significance

Abstract:Objective To investigate the expression of E- cadherin (E- cad), CD44v6 and matrix metal oproteinase- 2 (MMP2) in bladder urothelial carcinoma and its clinical significance. Methods The expression of E- cad, CD44v6 and MMP2 was detected by mmunohistochemistry Envision method in 105 specimens of bladder urothelial carcinoma and 20 specimens of normal bladder tissue. Results E- cad and CD44v6 were positive and MMP2 was negative in al 20 specimens of normal blad-der tissue. E- cad and CD44v6 were positively stained in 43.8%(46/105) and 58.1%(61/105)of bladder urothelial carcinoma specimens, respectively. The positive expression rate of MMP2 was 53.3%(56/105). There was a positive correlation between the expression of E- cad and CD44v6. The abnormal expression rates of E- cad, CD44v6 and positive expression rate of MMP2 were significantly correlated with the pathologic grades, clinical stages, high recurrence rate and poor survival. Conclusion The ex-pressions of E- cad, CD44v6 and MMP2 may be associated with malignant progression of bladder urothelial carcinoma and these proteins might be used as molecular markers for tumor progression, recurrence and prognosis.
