趋化性细胞因子受体CXCR4基因表达对人骨肉瘤细胞增殖和迁移的影响

摘    要:目的探讨趋化性细胞因子受体CXCR4基因表达对人骨肉瘤细胞增殖和迁移的影响。方法Real-time PCR 检测人骨肉瘤细胞株 HOS-8603、U2OS 和 MG-63 中 CXCR4 mRNA 的表达情况;构建、鉴定pcDNA3/CXCR4 重组质粒并将其转染到 HOS-8603 细胞中(PCDNA3/CXCR4 组),同时制备转染 pcDNA3 空质粒的阴性对照组(pcDNA3组)。MTT检测细胞增殖情况,穿膜小室重组细胞基底膜迁移实验检测细胞迁移情况。结果 HOS-8603细胞中CXCR4 mRNA相对表达含量低于U2OS、MG-63细胞(P 〈0.05)。转染后24、48h,pcDNA3/CXCR4 组 CXCR4 mRNA 表达量是 pcDNA3 组的 96 倍和 124 倍(P 〈0.05)。与 pcDNA3 组比较,转染后24、48、72 h,pcDNA3/CXCR4组细胞增殖率和发生迁移的细胞数明显增加(P 〈0.05)。结论 CXCR4的表达可能是促进骨肉瘤细胞增殖和迁移的重要因素。

Abstract:Objective To explore the influence of specific suppression of chemokine receptor CXCR4 gene on the proliferation and migration of human osteosarcoma cells.Methods Real-time PCR was used to detect the expression level of CXCR4 mRNA in human osteosarcoma cell lines HOS-8603,U2OS and MG-63.The recombinant plasmid pcDNA3/CXCR4 was transfected into the osteosarcoma cell HOS-8603 after the construction and identification(pcDNA3/CXCR4 group).At the same time,pcDNA3 empty plasmid transfection of the negative control group was preparected(pcDNA3 group).MTT assay was used to detect the effect of CXCR4 on cell proliferation.Transwell invasion chamber and reconstructed basement membrane examination were used to evaluate the effect of CXCR4 on cell migration.Results The expression level of CXCR4 mRNA inHOS-8603 cells was lower than those of U2OS cells and MG-63 cells(P 0.05).The expressions level of CXCR4 mRNA in pcDNA3/CXCR4 group were 96 times and 124 times higher than those in pcDNA3 group,respectively(P 0.05),at 24 h and 48 h after transfection of pcDNA3/CXCR4.The proliferation and migration of HOS-8603 cells transfected in pcDNA3/CXCR4 group were higher than those in pcDNA3 group(P 0.05),at 24,48,72 h after the transfection.Conclusion CXCR4 might be one of the important factors promoting the proliferation and migration of osteosarcoma cells.
