摘    要:目的:建立一测多评法(QAMS)同时测定复明片中红镰霉素-6-O-β-龙胆二糖苷、大车前苷、莫诺苷、决明子苷、马钱苷、山茱萸新苷和橙黄决明素-6-O-葡萄糖苷的含量。方法:采用Diamonsil C18色谱柱(250 mm×4.6 mm,5μm),柱温为30℃;检测波长为330 nm(大车前苷)、278 nm(红镰霉素-6-O-β-龙胆二糖苷、决明子苷和橙黄决明素-6-O-葡萄糖苷)和240 nm(莫诺苷、马钱苷和山茱萸新苷);流动相为乙腈-0.1%磷酸溶液,梯度洗脱,体积流量为1.0 mL·min-1。选取莫诺苷为内参物,建立该成分与其他6种成分的相对校正因子,计算各成分含量。将QAMS计算结果与外标法实测结果进行对比,验证所建立方法的可行性。结果:红镰霉素-6-O-β-龙胆二糖苷、大车前苷、莫诺苷、决明子苷、马钱苷、山茱萸新苷和橙黄决明素-6-O-葡萄糖苷分别在5.26~131.50、0.88~22.00、5.63~140.75、3.77~94.25、2.86~71.50、0.63~15.75、2.19~54.75μg·mL-1线性关系良好(r≥0.9993),平均加样回收率(RSD)分别为99.02%(1.21%)、97.43%(1.09%)、100.11%(0.64%)、98.49%(0.76%)、97.96%(1.19%)、96.98%(1.15%)、98.40%(1.51%),QAMS计算结果与外标法实测结果无明显差异。结论:该方法全面、简便、准确,可用于复明片中7种成分含量的同时测定及质量控制。

Abstract:Objective:To establish a quantitative analysis of multi-components by single marker(QAMS)method for the content determination of rubrofusarin-6-O-β-gentiobioside,plantamajoside,morroniside,cassiaside,loganin,cornuside I and aurantio-obtusin-6-O-β-D-glucoside in Fuming Tablets,simultaneously.Methods:The analysis was performed on a Diamonsil C18 column(250 mm×4.6 mm,5μm),the column temperature was set at 30℃.The detection wavelengths were set at 330 nm,278 nm and 240 nm.The mobile phase comprised of acetonitrile-0.1%phosphoric acid solution flowing at 1.0 mL·min-1 in gradient elution.Morroniside was selected as the reference substance and the relative correction factors of other six components were calculated.The feasibility of the method were validated by comparing the results of QAMS method with those of external standard method.Results:Rubrofusarin-6-O-β-D-gentiobioside,plantamajoside,morroniside,cassiaside,loganin,cornuside I and aurantio-obtusinβ-D-glucoside showed good linear relationships within the ranges of 5.26-131.50,0.88-22.00,5.63-140.75,3.77-94.25,2.86-71.50,0.63-15.75,2.19-54.75μg·mL-1(r≥0.9993),respectively,whose average recoveries(RSDs)were 99.02%(1.21%),97.43%(1.09%),100.11%(0.64%),98.49%(0.76%),97.96%(1.19%),96.98%(1.15%)and 98.40%(1.51%),respectively.The results obtained by QAMS approximated those obtained by external standard method.Conclusion:The method is comprehensive simple and accurate,which can be used for simultaneous the content determination and quality control of 7 components in Fuming Tablets.
