生 盐知母水煎液中芒果苷在大鼠体内的组织分布

摘    要:目的:考察生、盐知母水煎液中芒果苷在大鼠体内的组织分布情况,探讨盐知母引药入肾的作用机制。方法:将40只大鼠随机分为对照组、生知母(10 g·kg-1)组、盐知母(10 g·kg-1)组、芒果苷(200 mg·kg-1)组,灌胃给药2 h后处死,立即解剖采集心、肝、脾、肺、肾组织,采用高效液相色谱法(HPLC)测定组织样品中的芒果苷含量。结果:方法专属性、日内和日间精密度、稳定性及提取回收率均符合生物样品分析要求;各组织样本在检测浓度范围内呈良好线性关系;生、盐知母及芒果苷组大鼠给药2 h内芒果苷主要集中在肺、肾组织中,含量从高到低的顺序为肺>肾>脾>肝>心。结论:生、盐知母和芒果苷组大鼠体内芒果苷均集中分布在肺、肾组织中,与知母的归经特点相符。生、盐知母组大鼠体内芒果苷在肺、肾组织中的含量明显高于芒果苷组,但生、盐知母组大鼠各组织中芒果苷含量差异无统计学意义。

Abstract:Objective:To investigate the tissue distribution of mangiferin in rats after the administration of Anemarrhenae Rhizoma and salt stir-fried Anemarrhenae Rhizoma decoction,explore the mechanism of salt stir-fried Anemarrhenae Rhizoma leading drugs into the kidney meridian.Methods:40 Rats were randomly divided into control group,Anemarrhenae Rhizoma group(10 g·kg-1),salt stir-fried Anemarrhenae Rhizoma group(10 g·kg-1)and mangiferin group(200 mg·kg-1).The rats were executed 2 hours after intragastric administration,and immidiately dissected.The heart,liver,spleen,lung and kidney were collected and the content of mangiferin in the tissues were determined by high performance liquid chromatography(HPLC).Results:The method′s specificity,intra-day and intra-day precision,stability,and extraction recovery met the requirements of biological sample analysis.The tissue samples showed a good linear relationship in the detection concentration range.Mangiferin was mainly concentrated in the lung and kidney within 2 hours after administration in groups of Anemarrhena Rhizoma,salt stir-fried Anemarrhenae Rhizoma and mangiferin.The order of mangiferin content from high to low was lung>kidney>spleen>liver≥heart.Conclusion:Mangiferin of three groups is mainly concentrated in lung and kidney,which is consistent with the meridian tropism of Anemarrhenae Rhizoma.The content of mangiferin in lung and kidney tissues of Anemarrhenae Rhizoma group and salt stir-fried Anemarrhenae Rhizoma group is significantly higher than that of mangiferin group.However,there is no statistical difference between Anemarrhenae Rhizoma group and salt stir-fried Anemarrhenae Rhizoma group.
