Abstract:Objective To screen the key genes that affect the occurrence and development of ovarian cancer and to analyze their mechanisms of action.Methods The key genes of ovarian cancer were screened by using Gene Expression Omnibus(GEO),the Cancer Genome Atlas(TCGA)and Kaplan-Meier Plotter(KMplot)databases,and their relationship with the patients′clinicopathological characteristics was analyzed.At the same time,GESA was used for cluster analysis of signal pathways,and the TIMER database was used to identify the genes closely related to it.The interaction network was mapped through the STRING database to predict its mechanism of action.Results A total of 13 differentially expressed genes were obtained through screening,among which Aristaless Related Homeobox(ARX)and Family with Sequence Similarity 150 Member B(FAM150B)were related to the prognosis of the patients,and the lower their expression levels,the shorter the overall survival time of the patients(P<0.01).Further analysis found that the expression levels of ARX and FAM150B were also related to the expression levels of interleukin-6(IL-6),Phosphoinositide-3-Kinase Regulatory Subunit 2(PIK3R2),PIK3CD,VAV1,PIK3CA,RAC2,PIK3R1,and MAPK1(P<0.05).The analysis of STRING database showed that ARX and FAM150B regulated the above genes through TUBA1A,EGFR and PLXNA2.Conclusion The expression levels of ARX and FAM150B is low in ovarian cancer tissues,and their expression levels are closely related to the prognosis of the patients.ARX and FAM150B can regulate a series of genes,thereby weakening the body′s anti-tumor immune response and thus promoting the occurrence and development of ovarian cancer.
