Abstract:Objective To develop a ultra-high performance liquid chromatography tandem mass spectrometry method for the quantitative analysis of 30 phenolic compounds in Pholidota chinensis Lindl.(P.chinensis),and 13 batches of P.chinensis samples from different areas were evaluated.Methods Chromatographic separation was performed on Welch Xtimate UHPLC C 18(2.1 mm×100 mm,1.8μm)with mobile phase consisting of 0.1%formic acid solution and acetonitrile at a flow rate of 0.2 mL·min-1 using gradient elution,and the column temperature was 45℃.MS monitoring was performed by electrospray ionization in positive and negative ion switching mode using multiple reaction monitoring;SPSS software was employed to perform the hierarchical cluster analysis for the content determination results.Results The calibration curves for 30 compounds had good linearity(r≥0.9986)in their linear range.The average recoveries were in the range of 95.18%~104.97%(RSD≤5.04%),the RSD of precision,repeatability,and stability were all less than 5.19%.Hierarchical cluster analysis classified 13 batches of P.chinensis from different areas into 2 categories,including the medicinal materials from Zhejiang,Fujian,Guangdong and Guangxi(S1~S3,S5~S11,S13)asⅠcategory,while those from Sichuan and Yunnan(S4,S12)asⅡcategory.Conclusion The method developed in this study can quickly achieve qualitative and quantitative detection of 30 phenolic compounds in P.chinensis,which can be used in the quality evaluation of P.chinensis.
