| 摘 要： | 目的：研究发现脑缺血患者体内存在N甲基－D－门冬氨酸（NMDA）受体自身抗体，了解共滴度与疾病程度的相关性。方法：用NMDAR1单克隆抗体淘筛噬菌体展示随机12肽库，获得NMDR1模拟表位抗原。结果：细胞竞争ELIA结果显示，该模拟抗原可以竞争细胞表达天然抗原与抗体的结合，具有抗原模拟性。结论：噬菌体肽库中筛选NMDAR1受体模拟抗原可以用于临床病人血清检测，本研究为下一步临床检验准备了必要条件。 |

| Abstract: | To determine the minotope of NMDAR1. The monoclonal antibody against the NMDAR1 was used as target protein to screen the binding peptide from a 12-mer Ph.D. random peptide library. After three rounds of affinity screening,the peptide sequences of positive phage clones were determined and analyzed by DNA sequencing,ELISA and competitive inhibition. A positve clone was found. It could competitively inhibit the native antigen binding to mAbN1. [Conclusion] This peptide mimics native NMDAR1 and may be useful for NMDAR1 autoantibody diagnosis in the patients with chronic cerebral circulation disorder. |
