滋补脾阴方药对脾阴虚糖尿病大鼠海马内质网应激影响的研究

摘    要:目的:探讨滋补脾阴方药(ZiBu PiYin Recipe,ZBPYR)对脾阴虚糖尿病大鼠海马内质网应激相关分子表达的影响。方法:高脂饲料喂养联合小剂量链脲佐菌素(streptozocin,STZ)注射建立2型糖尿病模型,在此基础上,通过复合因素造模的方法建立脾阴虚糖尿病模型,以水迷宫实验评价模型大鼠是否出现学习记忆障碍,之后观察各组大鼠海马内质网应激相关分子葡萄糖调节蛋白78(glucose regulated protein 78,GRP78)、蛋白激酶样内质网激酶(protein kinase RNA-like endoplasmic reticulum kinase,PERK)和真核转录因子2的α亚单位(eukaryotic translation initiation factor 2 subunitα,eIF2α)表达的变化。结果:ZBPYR治疗后GRP78 mRNA水平降低(P0.05),GRP78、磷酸化PERK、磷酸化eif2α蛋白水平明显降低(P0.05),与脾阴虚糖尿病组相比,差异有统计学意义。结论:滋补脾阴方药可能是通过影响内质网应激PERK信号传导改善脾阴虚糖尿病大鼠学习记忆障碍。

Abstract:Objective:To investigate the effects of Zibu Piyin Recipe(ZBPYR)on endoplasmic reticulum stress(ERS)-related protein expression in hippocampus of Spleen-Yin deficiency diabetic rats.Methods:Type 2 diabetes model was established by high fat diet combined with low-dose streptozotocin(STZ)injection.Then we induced spleen-yin deficiency diabetic model by defatigation,hungry-full diet and drenching the drug of damaged yin.To evaluate whether diabetic rats and spleen-yin deficiency diabetic rats were impaired in learning and memory,we performed Morris Water Maze test.We detected the expressions of GRP78,PERK and eIF2,which were related with ERS in cortex of rats.Results:After treating with ZBPYR,the protein levels of GRP78,phosphorylated PERK and phosphorylated eIF2 and the mRNA expression of GRP78 were significantly decreased compared with Spleen-Yin deficiency diabetic rats.Conclusion:ZBPYR may improve learning and memory impairment in Spleen-Yin deficiency diabetic rats by affecting the endoplasmic reticulum stress PERK signaling.
