| 摘 要： | 目的:研究中药蛇六谷石油醚萃取物诱导人胃癌SGC-7901细胞凋亡的机制。方法:以MTT法检测蛇六谷石油醚萃取物对人胃癌SGC-7901细胞增殖的影响;Western Blot法检测细胞凋亡相关蛋白Survivin、Bcl-2、Bax、Cleaved-Caspase-9的表达。结果:该提取物对人胃癌SGC-7901细胞增殖存在明显的抑制作用,且呈剂量-效应关系。Western Blot实验结果显示该提取物能明显下调Survivin蛋白、Bcl-2蛋白的表达,上调Bax蛋白、Cleaved-Caspase-9蛋白酶的表达;结论:该提取物诱导SGC-7901细胞凋亡的机制可能与影响上述蛋白的表达。

| Abstract: | Objective:To study the underlying principles of cell apoptosis which caused by Rhizoma amorphophalli with human gastric cancer cells SGC-7901.Method:Test the effect caused by petroleum ether extract of Rhizoma amorphophalli on gastric cancer SGC-7901 cells proliferation with MTT.Take different concentrations of petroleum ether extract of Rhizoma amorphophalli acting on SGC-7901 cells.Analyze the expressions of Survivin,Bcl-2,Bax and Cleaved-Caspase-9 with Western Blot method.Results:Different concentrations of petroleum ether extract of Rhizoma amorphophalli had diffierent inhibiting effect on SGC-7901 cells proliferation.With the increase of concentration,the inhibition gradually enhanced.After 24 hours’ acting on the SGC-7901 cells,all of different concentrations of petroleum ether extract of Rhizoma amorphophalli can obviously down-regulate the expressions of Survivin protein and Bc1-2 protein.At the same time it can obviously up-regulated the expressions of Bax protein and Cleaved-Caspase-9 protein.Conclusion:The possible mechanism that make SGC-7901 cells apoptosis of petroleum ether extract of Rhizoma amorphophalli may be related to the influence on the expressions of above proteins. |
